This bachelor thesis deals with the evaluation of the energy performance of the Mars hotel in Špindlerův Mlýn. The theoretical part of this thesis is focused on the conversion of sunlight into electric power using solar-powered photovoltaics. The practical part is focused on the analysis of energy consumption and energy flux of the inspected building. The final part C concerns about the result of this thesis – the energy performance certificate of the analyzed building.

Economical office building

The main aim this master's project is to design a nearly zero-energy office building. The project has three parts. Design of building itself and building services in the first and second part. Third part is specialized. The low-rise building has three storeys. On the first floor are small offices under 30 square meters and technical facilities of building. On the second floor are larger offices over 30 square meters and some staff area. On the third floor are one large shared office and local library. All walls are designed from ceramic blocks and external walls are insulated by ETICS. The floor slab and flat roof are designed from reinforced concrete. External look is from natural materials to fit the local environment. The heating to the building is delivered by condensing gas boiler. The next service systems in the building are ventilating, lighting and air conditioning. The source of renewable energy is photovoltaics. In the third part I proposed different types of structures and heating sources and I used them in the Energy Performance Certificate.

Konstrukce neoznačených mutantů v genech kódujících Ser/Thr proteinkinasy a fosfoproteinfosfatasu Pseudomonas aeruginosa a analýza jejich vlastností

To investigate the importance of individual protein kinases and phosphatases in P. aeruginosa physiology we prepared the single and double mutants in the coresponding genes. Single mutants in protein kinase genes stk1, ppkB and protein phosphatase gene stp1 were prepared by the method of gene replacement with subsequent excision of antibiotic cassettes using Flp-FRT recombination system. Two-dimensional gel electrophoresis was used to analyze the patterns of phosphoproteins obtained in in vitro protein kinase assays with the cell-free lysates of wild type as well as all mutant strains.
