Synchronization of circadian clock in hippocampus
Kubátová, Eliška ; Sumová, Alena (advisor) ; Liška, Karolína (referee)
Circadian rhythms refer to the physiological, biochemical and behavioral changes of the organism with a period of about 24 hours, such as the cycle of sleep and wakefulness, the release of hormones, enzymatic activity or gene expression. Apart from the central pacemaker located in the suprachiasmatic nucleus (SCN) of the hypothalamus, we find many other peripheral oscillators in which we observe the rhythmic expression of clock genes. In this study, we focused on one of the peripheral brain oscillators located in the hippocampus. The hippocampus is best known for its role in memory formation through the mechanism of long- term potentiation (LTP). The formation of LTP is another process that exhibits circadian variation. Although the rhythmic expression of genes in the hippocampus has already been demonstrated, the mechanisms of synchronization of these clocks remain a subject of investigation. In this study, we studied the effect of candidate substances - N-methyl-D- aspartate (NMDA) and leptin on the circadian clock of the hippocampus. Genetically modified mPer2Luc mice were used as a model. The main goal of the work was to introduce a method of preparing organotypic explants of the hippocampus of these mice for monitoring the circadian clock in real time using the LumiCycle device. Next goal was...
Epigenetic mechanisms in psychostimulant addiction
Kubátová, Eliška ; Novotný, Jiří (advisor) ; Rydzyková, Tereza (referee)
Psychostimulants are highly addictive substances that act in a brain reward center by increasing extracellular levels of monoamines. In western civilization, they have gained popularity during the 20th century. Psychostimulant addiction is a persistent social problem even today since therapy is often unsuccessful and former users relapse frequently. Alterations in gene expression occur after psychostimulant is administered. These changes are long-lasting as they can often persist for weeks and months. This work focuses on the epigenetic changes in a brain associated with the development of addiction after psychostimulant administration. Key words: Epigenetics, psychostimulants, amphetamine, methamphetamine, cocaine, addiction
The concept of humanitarian intervention from the Russian perspective
Kubátová, Eliška ; Střítecký, Vít (advisor) ; Karlas, Jan (referee)
The diploma thesis is concerned with the approach of Russian federation towards the humanitarian intervention concept, respectively to the responsibility to protect (R2P) doctrine, which was endorsed by all member states of the United Nations at the 2005 World Summit. The main idea of the doctrine is a duty of all states to protect their citizens, and if certain state is manifestly failing to protect its population, the international community has responsibility to use appropriate measures in order to protect lives of civilians. Therefore in extreme cases, states have the right to military intervene on a territory of other state. The Russian federation, which traditionally promotes principles of state sovereignty and non- interference, rejects the liberal conception of the humanitarian intervention and strongly criticizes interventions led by the West. On the other hand, Russia uses the concept if its own interests are at stake. The thesis illustrates this ambiguous perception of the humanitarian intervention concept by the Russian federation on several historic events, when Moscow either criticized Western states' implementation of the R2P concept (NATO intervention in Kosovo or the cases of Libya and Syria), or used the concept on its own in order to legitimize military intervention (Chechnya,...
