Alcohols utilization as non - freezing heat transfer liquids
Kořínková, Klára ; Bednařík, Karel (referee) ; Kotlík, Josef (advisor)
This bachelor´s thesis deals with the alcohols used by the non-freezing heat transfer fluid. Were compared alcohols methanol, ethanol, ethane-1,2-diol, propane-1,2-diol and propane-1,3-diol. The substances were examined mainly from the toxicological point of view and negative impacts on human health. Toxic substance was determined by ethane-1,2-diol. Propane-1,2-diol is the most used substance in the market which is non-toxic.
Ecotoxicity of selected platinum metals.
Kořínková, Klára ; Vávrová, Milada (referee) ; Zlámalová Gargošová, Helena (advisor)
Over the last 20 years increasing concentration of platinum metals in the environment was recorded. These hazardous metals come through anthropogenic activities in the aquatic and terrestrial ecosystem. It is therefore necessary to deal with their possible negative impact on organisms. This thesis is focused on the ecotoxicological evaluation of selected complex compounds of platinum metals, namely platinum and palladium. For ecotoxicity evaluation tests on following aquatic organisms were used: Daphnia manga, Thamnocephalus platyurus and Lemna minor. On the base of obtained results ecotoxicological values of LC50 and EC50 were calculated.
Role of PIM oncogenes in the biology and chemoresistance of aggressive lymphomas
Kořínková, Klára ; Klener, Pavel (advisor) ; Petrák, Jiří (referee)
Proviral integration site for Moloney murine leukemia virus (PIM) kinases are serine/threonine kinases and oncoproteins involved in tumorigenesis of many solid and hematopoietic malignancies including mantle cell lymphoma (MCL) and diffuse-large B-cell lymphoma (DLBCL). They were shown to promote growth and survival of cancer cells by phosphorylation of proteins involved in cell cycle regulation, transcription, translation and apoptosis. Their potential as therapeutic target is, however, complicated by existence of overlapping signaling pathways. Here we show that inhibition of PIM kinases with AZD1208, highly selective pan-PIM inhibitor, reduces growth of cell lines derived from MCL and DLBCL patients. Inhibition of PIM kinases results in decreased phosphorylation of proteins involved in apoptosis and cap-dependent translation. We further showed that concurrent inhibition of PIM-kinases with AZD1208 and signaling from B-cell receptor with ibrutinib (a Bruton's tyrosine kinase (BTK) inhibitor) or idelalisib (a phosphatidylinositol 3-kinase (PI3K) inhibitor) synergistically reduces growth of MCL cell lines. Combination of AZD1208 and ibrutinib was accompanied by decreased phosphorylation of eukaryotic translation initiation factor 4E binding protein 1 (4E-BP1) and decreased expression of...
Molecular mechanisms of G2/M checkpoint regulation
Kořínková, Klára ; Macůrek, Libor (advisor) ; Forman, Martin (referee)
Cell division is necessary for maintaining tissue homoeostasis, but at the same time its defects are closely related to the development of many diseases including cancer and premature ageing. Activation of oncogenes leads to replication stress and directly threatens genome stability. The right control of transition between interphase and mitosis is an important mechanism for the protection of genome integrity. Nuclear division is only possible with those cells in which flawless duplication of genetic information occurred. By contrast, cells with damaged DNA structure remain temporarily or permanently stopped at G2 phase of the cell cycle. The topic of this thesis is a detailed literature overview with the subject of molecular mechanisms of the G2/M transition regulation under unperturbed conditions and in the presence of damaged DNA.
