Development of methodology for detection of circulating nucleic acids in cancer patients

Cancer diagnosis represents one of the most common causes of death in the world, also linked to an ever-increasing incidence of even some more difficult-to-treat variants. One of the most common types of cancer in the world is colorectal cancer, which ranks second in terms of mortality. For these reasons, various methods have been developed for an early diagnosis of the disease. One of the modern and non-invasive methods is the so-called liquid biopsy, based on the detection of circulating DNA and RNA from the blood of patients. This method is also suitable for choosing a therapy, monitoring its effectiveness, and observing the relapse of the disease. In this work, a methodical workflow was created for the monitoring of circulating nucleic acids for its further use in a cohort of patients with colorectal cancer. Various commercial kits were used for the isolation of circulating nucleic acids from plasma of healthy controls, among which the best ones, in terms of quality and quantity, was chosen. The threshold for hemolysis detection by qPCR was also determined using samples from healthy donors. Study of plasma discharge effect on distilled water pH

This tesis is concerned with the influence of Gliding arc plasma discharge on the pH of distilled water while using different treatment time and distance between Gliding arc and the surface of distilled water. The theoretical part consists of research into various studies, whose main focus is the practical usage of Gliding arc and the possibilities of its use. The practical part contains the values of the measured quantities, models and graphs illustrating the effects of the process. The conclusion offers a brief summary of the findings and claims, whose correctness needs to be verified in further studies.
Preparation of transcription factor FOXK2 - DNA binding domain

Transcription factors are specific proteins, which are able to regulate transcription and which are divided into individual families based on structural motifs. One of these families is the FOX family, which also includes the transcription factor FOXK2, which was the studying subject of this work. The FOXK2 protein regulates many cellular processes, including cell proliferation and metabolism and plays an important role in carcinogenesis, as its high expression has been reported in cases of lung, liver and kidney cancer. In this work, the DNA binding domain of the transcription factor FOXK2 was prepared by recombinant expression of Escherichia coli cells. The obtained protein was afterwards purified and its interaction with DNA was studied by native mass spectrometry.

The bachelor thesis concerns itself with plasma discharge Glidarc influence on choosen water parameters, like density, surface tension, dynamic viscosity, heat capacity and pH. The theoretical part is a brief introduction to plasma and gas-discharge matters. The practical part contains measurment outcome and corresponding calculations. The conclusion includes a summary and evaluation of the whole research.
Aestethic Education as a New Subject at Pouchov Primary School

The Diploma thesis with the possible implementations of Aesthetic education from 6th to 9th grades of primary school. The subject matter concerns all fields befalling into the educational area of Arts and Culture, as defined by the Framework Educational Programme for Basic Education; music, art, drama education, film/audiovisual education, and dance and movement education. In an educational approach Aesthetic education as a subject is based on artephiletics. In addition to giving the theoretical grounds for creating the subject, the dissertation also offers an account of the preparatory phase of its implementation at a specific primary school, including inspirational materials for teaching the subject from 6th to 9th grade at this school.
