Production of PHB-HV from grape pomace by employing a pure culture of Cupriavidus necator

This bachelor thesis deal with the production of PHB-HV copolymer employing a pure culture of Cupriavidus necator H16. First, Erlenmeyer flask´s cultivations using pure fructose as a source of carbon were performed. The mode of addition of valeric acid for the synthesis of PHB-HV was studied. Furthermore, the effect of stress factor (ethanol) on PHB-HV synthesis by this microorganism was investigated. The method obtained by optimization in Erlenmeyer flasks was applied to the synthesis of the PHB-HV copolymer in the bioreactor using fructose as a source of carbon, too. Furthermore, after enzymatic hydrolysis, a sugar extract from grape pomace was obtained as a source of carbon for the synthesis of PHB-HV. The fermentation in Erlenmeyer flasks and the bioreactor using an extract of grape pomace was carried out. Using this alternative carbon source 3.51 g/l PHB-HV copolymer was produced in Erlenmeyer flask, 5.06 g/l in bioreactor respectively. In the first case, the proportion of 3-hydroxyvalerate in the copolymer was 23.78 mol. % and in the second one, 9.39 mol. %.
Development of microbioreactor for microbial biotechnology

This master thesis deals with the development of a minibioreactor for microbial biotechnologies. The AlgaTox system – an analytical photobioreactor from BVT Technologies – was the default unit of the new device. The working volume of the new minibioreactor is in the range of 4 to 8 ml. The minibioreactor was composed of a minithermostat, a reaction vessel, an oxygen electrode, a temperature and pH probe, accessories for the supply of air to the liquid and an insertion for the transport of liquid from / to the reactor. The functionality of the assembled device and its characteristics were measured. Furthermore, an operating procedure for the decontamination process using a hydrogen peroxide mist was developed for the new equipment. An operating procedure for culturing microorganisms in a minibioreactor was also set up. Pilot cultivations of Halomonas halophila were demonstrated in the prepared equipment. In these cultivations, three assemblies for air supply to the liquid were tested. However, none of the assemblies was able to provide a sufficient supply of oxygen to cell culture – the dissolved oxygen value always dropped to 0%. For the further development, a new assembly was designed for the supply of air to the liquid - an aeration ring from a membrane tube.

Woman on The Labour Market in the Situation of the Mother

Human society is dependent on women who ensure their sustainability in the role of mothers. At the same time women participate in the economic prosperity of the family and society. The combination of these two roles puts them at a disadvantage in the labour market. This thesis aims at analyzing the current situation of women from the point of view of motherhood and position on the labour market. The theoretical part deals with two primary areas including social policy and the labour market. Social policy gives information about family policy and state support to mothers in comparison with other countries. The practical part, which is based on methodology, is dedicated to the analysis of the current situation of women and is divided according to the origin of the data. The secondary data were obtained from the public statistical database and the primary one through the questionnaire survey. The outcome of the work reveals significant factors influencing the age at which women have their first child, the dependence between parental leave income, the length of parental leave, and other answers to the research questions.
Tourist Product in the South Bohemian Region

This bachelor thesis analyses the current offering and assumptions of tourism in the city of Jindřichův Hradec. Based on this analysis, thesis aims to propose a new potential tourist product which is in line with current trends. The product is constructed to increase short duration stays in the city. Short stays and low number of spent nights overall were the main reasons for choosing the city of Jindřichův Hradce, out of a tourism region of Jižní Čechy. The goal is achieved while using methods of comparison, analysis, and synthesis. In the last chapter there is an introduction to a tourist product called Za poznáním Jindřichohradecka na kole.
