Development and implementation of new approaches for proteomic characterization of bone tissues in oral surgery

1 Abstract Proteomics is a booming field with application in many areas of medicine, including dentistry. Nevertheless, proteomic characterization of bone tissues in oral surgery is not still commonly used. The main reason is involvement of demanding analytical approaches due to insoluble chatacter of bone tissues. The goal of this work was to develop and apply straightforward methodology that could lead to the routine use of proteomics in this area as well. Using porcine jawbones as a model samples, a technique was developed allowing identifying about hundreds of proteins thanks to their trypsin digestion directly in bone tissues ("in-bone digestion") followed by analysis using liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). This technique was subsequently applied to the analysis of human maxillary and mandibular bone tissues obtained during surgical procedures. In both maxillary and mandibular bone samples, it was possible to identify a considerable amount of proteins using the "in-bone digestion" technique. Analysis of organic additives in historical mortars

- 4 - Abstrakt The aim of this diploma thesis was to determine the possibility of identifying the protein and lipid additives, such as blood, gelatine, curd, lard and eggs, in the model samples of lime based mortars using Raman spectroscopy and gas chromatography- mass spectrometry. It was proved that the method of Raman spectroscopy can determine the presence of organic additives in the order of one weight percent. The gas chromatography enables to detect lipid additives from the order of hundredths weight percent. Unambiguous resolution of protein additives can be done in the samples with the concentrations up to the order of one weight percent of the additives.
Protein composition of flours and bakery products

The aim of this study was to carry out an analysis of a protein composition of diferent crop species using the method of peptide mass mapping. The samples were analysed using analytical method LC/ESI-Q-TOF and the acquired spectra were compared with the plant protein databases. The origin of the found plants was discussed in the aspect of the reliability of the used method considering the taxonomical relationships in the taxonomical group. The results of the analysis were compared with the food composition specified by the producer, and it was evaluated, if it is possible to verify the protein composition by using the applied method. In case of maize (Zea mays) this method is considered as reliable, because all of the found proteins were indentified as maize proteins. In case of rice (Oryza sativa) and pea (Pisum sativum), only one, respectively, two proteins of different plants species were found in the number of the relevant plant proteins. This method was evaluated as not so reliable for samples made of cereals (Poaceae). It is possible to conclude, whether the product comes from Poaceae or not, but it is impossible to verify the taxon of genus or the inferior taxons. Application of vibrational spectroscopy to analysis of artwork - mortars

The aim of this work was to determine the possibility of identifying the protein and lipid additives, such as blood, gelatin, curd, lard or eggs, in the model samples of lime based mortars using infrared spectroscopy. We have focused on the reflectance techniques of infrared spectroscopy, and we proved that by these methods can be determined the presence of additives, down to tenths percent by weight. Detailed resolution of lipid and protein additives can be achieved only for samples containing at least one weight percent of additive. There were not observed any spectral changes which can be correlated with performed curing and aging of prepared model samples.
Iron gall ink

This work is focused on history and preparation of gall-nut ink - the most common agens of writing for more than eight centuries. This thesis contains information about each component of the ink and explains the reason why it was added to the ink. The work also explains the chemical principles, that are responsible for the formation of the pigment and for the behaviour of the mixture when it is applied on the substrate and exposed to atmospheric oxygen. Experimental part of this work focuses on practical preparation of two gall-nut inks by medieval recipes and their quality tests.
The study of influence of inorganic pigments on the identification of egg tempera by mass spectrometry

The aim of this diploma thesis is to find out in what way inorganic pigments influence the identification of egg and yolk tempera using mass spectrometry MALDITOF (matrix-assisted laser desorption/ionization time of flight). The other aim was to prepare a set of model color layers including ten inorganic pigments combined with the egg and yolk tempera. Using the method of peptide mass fingerprinting mass spectra of the samples were found out and the obtained peptides were compared. Finally the egg tempera was distinguished from the yolk tempera and it was found which pigments negatively influenced the identification of the temperas. The m/z values were found out for both temperas and they were used to upgrade database of the reference proteinaceous binders that have been used in art works.

The goal of the thesis is to find out the value of the company ČSAD Česká Lípa a. s. The thesis is dividend into two parts, scilicet a theoretical part and a practical part. The theoretical part explains particular analyses, valuation procedure and methods used in the practical part. It is necessary to make strategic and financial analysis before valuation of the company. These analyses help to build a financial plan, which is used as a basis for valuation of the company by income method. A discounted cash flow method is applied for the valuation of the company, specifically the two-phased model using free cash flow to firm.
