The main aim of this master´s thesis is design and Cheb load-carrying steel structure of railway station in two variatons. A given object is situated in Ústí nad Orlicí.The steel structure is created by central nave, his dimensions is 30,0 x 30,0 m, and two secondary naves and his dimensions is 18,0 x 18,0 m.The central nave is two-storey height 12,4 m and secondary nave is single-storey height 5,5 m. Carrying building systém is created croos-column links, which are composed pipes. Cross-links are saved on fixed columns. The calculation was done through the use of Scia Engineer 2008 program and by myself alone.

Lukášková, M.: Higher plants tissue cultures 28. Diploma thesis, Charles University in Prague, Faculty of Pharmacy in Hradec Králové, 2007. A biotransformation possibility of the tissue culture Centella asiatica (L.) was studied. These cultures were added the precursors of arbutin: hydoquinone, tyrosine, 4-hydroxybenzoic acid and p-coumaric acid into the medium. The precursors were used in a concetration of 100 mg/l and the period of their action was 6, 12, 24, 48 and 168 hours. Positive results (both TLC and HPLC) in arbutin production were obtained in the suspension culture after an addition of hydroquinone. The largest amount of arbutin was achieved after a week's cultivation with hydroquinone (0,0429%).

Thesis, Charles University in Prague, Faculty of Pharmacy in Hradec Kralove, Department of Botanic and Environmental Science, Pharmacy (2010) This paper focused on the study of biotransformation activity of suspension culture Datura meteloides DC. ex Dunal after addition of arbutin exogenous precursor hydroquinone into nutrient medium. The percentage of arbutin content was tested depending on the type and concentration of growth regulator in nutrient medium (IAA, IBA - concentrations of 0,1; 1,0; 10,0 mg/l), concentration of arbutin precursor hydroquinone (100 and 200 mg/l) and intervals between collection of samples for analysis, i.e. cultivation time with precursor (24, 48, 168 hours). Culture transformed hydroquinone into arbutin regardless of the type and concentration of growth regulator, precursor concentration and the length of the experiment. The highest percentage of arbutin content in culture extract (8,11 %) was detected after 24 hour cultivation using hydroquinone in concentration of 200 mg/l and growth regulator IAA in concentration of 0,1 mg/l. Diplom Paper, Charles University in Prague, Faculty of Pharmacy in Hradec Králové, Department of Pharmaceutical Botany and Ecology, 2007, p. 62 This work aimed at biotransformation of exogenous precursor of arbutin - hydroquinone with in vitro cultures of Datura meteloides D. C. ex Dunal. Into the medium was added precursor of arbutin hydroquinone in the concetrations 100 mg/ l and 200 mg/ l. The cull was operated after 24, 48, 72, 96, 120, 144, 168 hours (1 week). The cultivationes of calluses prosecuted in the light, on the liquid medium MS [10] with addition 2, 4-diclorfenoxyacetic acid (1ml/ l) on the filter - paper bridge. Positiove resultes was obtained (TLC and HPLC analysis) in produktion of arbutin in all bridging cultures in both concetrationes (100 mg/ l, 200 mg/ l). The concentration of arbutin increased with the period of cultivation by all concetrationes of hydroquinone. The biggist value arbutin by concentration of hydroquinone 100 mg/ l was obtained by period of cultivation 168 hours, it was 0,41 %. By concentration of precursor 200 mg/ l, maximum value of arbutin 1,09 % was obtained after the biggest period of cultivation 168 hours.

Lucie Strnadová Microscopy of herbal drugs in Pharmacopoeia Bohemica MMIX. II Thesis Charles University in Prague, Pharmaceutical Faculty in Hradec Králové Field of study Pharmacy Microscopy is one of the identity exam of herbal drugs. The aim of the task is preparation of permanent preparatives of the drug called Sanguisorbae radix, description of its anatomical structure and furthermore process the image documentation of identifying characteristics of powder drugs' wet preparations formed by roots and rhizomes. Permanent microscopical preparations were made by procedure of embedding in paraffin, coloring was made by haematoxilin and safranine. Powder drugs' microscopical preparations were proceed by dispensatory in pharmacopoeia. Photodocumentation was made by using programme called Quick Photo Micro 2.3. The precursors of arbutin were added into the medium (hydroquinone, tyrosine, p-coumaric acid, hydroxybenzoic acid). The precursors were used in a concentration of 200 mg/l and the period of their action was 6, 12, 24, 48 and 168 hours (1 week). The cultivation of calluses was created by two different ways - by suspension culture and by culture cultivated on the filter-paper bridge. The cultivation was prosecuted on the light. Positive results (both TLC and HPLC analysis) in arbutin production were obtained in all suspension cultures after an addition of hydroquinone. The largest amount of arbutin in callus cultures was measured after a 48 hours cultivation (0,043 %). After a 168 hours cultivation was amount of arbutin only 0,038 %. In this experimental variant arbutin was also released into the medium (0,096 %). Biotransformation experiments with the others precursors of arbutin were not successful. The way of cultivation by the filter-paper bridges was also unsuccessful.

Diploma thesis, Charles University in Prague, Faculty of Pharmacy in Hradec Králové, 2008 The goal ofmythesis was to verify biotransformation posibility of theDatura meteloides culture. Hydroquine - the precursor of arbutin was added into these cultures. Precursor used in this study was prepared in concentration of 100 mg/l and 200 mg/l. Exposition of precursor was 24, 48, 72, 96, 120, 144 and 168 hours under constant conditions. By the qualitative and quantitative analysis I found the presence of studyied arbutin metabolite in all observed samples. The biggest arbutin volume was obtained after 168 hours of cultivation with precursor of concentration at 200 mg/l ( 3,05%). The in_uence of precursor's concentration and exposition time to the _nal product was thereby con_rmed.

An Abstract: Evaluation is a crucial part of every human activity and every qualified school teacher knows that it is not possible to do the work without evaluating activities. That's why the teacher's evaluation becomes a necessary, meaningful and obvious part of the educational process. It belongs to the most difficult teaching activities. The target of my bachelor's work is to find out teachers' attitudes and practical experiences with evaluation. In a detailed analysis I tried to show the extensive and wide range that the ability contents. The meaning of my work is to describe and analyse all the evaluating activities on the basis of a qualitative research using a method of an interview. I tried to find out in which different professional contents nursery school teachers and manager school teachers do their evaluating activities. The work is divided in a theoretical and a practical part. The fisrt part is a summary of evaluation theoretical knowledge based on a study of technical literature. In the bachelor's work there are described processes of teacher's evaluating activities. In the practical part there are mentioned the results of the research which was carried out by a method of an interview with three nursery school and basic school teachers.
