Role of Psb28 proteins in the biogenesis of the Photosystem II complex in the cyanobacterium Synechocystis sp. PCC 6803

The thesis focuses on the role of Psb28 proteins, namely the Psb28-1 and its homolog Psb28-2, in the biogenesis of the Photosystem II complex (PSII) in the cyanobacterium Synechocystis PCC 6803. The aims of this work were to localize the proteins within the cells, and to determine their function. A fraction of both Psb28 proteins was identified in the monomeric PSII core complexes but most proteins were found in the unassembled protein fraction associated with thylakoid membranes. Psb28-1 was mostly detected as a dimer while Psb28-2 as a monomer. Psb28-1 also differed from Psb28-2 by its higher affinity to the PSII core complex lacking CP43 antenna. Characterization of Psb28-less mutants suggested regulatory function of the proteins in PSII biogenesis in connection with chlorophyll biosynthetic pathway. Analysis of preparations isolated using FLAG-tagged versions of Psb28 proteins showed their association with Photosystem II - Photosystem I supercomplexes, especially under increased irradiance, and supported a role of Photosystem I in the PSII biogenesis.
Identification and sequencing genom of a new virus infecting lucerne

Samples of lucerne plants characteristic with local necrosic lesions, leave malformation and yellow spots on leaves were investigated with transmission electron microscopy. Virus particles observed there were filamentous ones of 600 to 700 nm long. Nucleic acid was isolated, transcribed and amplified using PCR. Genus-specific primers were designed based on reverse genetics from the highly conserved genes for carlaviruses, potexviruses and potyviruses. Successful amplification with carlavirus-specific primers, sequencing and comparison with sequences in GenBank database revealed presence of a carlavirus. This was later identified by nucleotide sequence comparison as a new isolate V4 of Alfalfa latent virus. Specific primers for isolate V4 were designed in a coat protein position. Half of the genom of this virus was obtained with PCR and PCR modified amplifications and compared with sequences of Alfalfa latent virus and Pea streak virus from GenBank.
Identification of filamentous virus which infected red clover (\kur{Trifolium pratense})

Samples of red clover plants characteristic with dwarf growth were investigated with transmission electron microscopy. Most frequent virus particles observed there were filamentous ones of 300 to 800 nm long. Nucleic acid was isolated, transcribed and amplified using PCR with potexvirus- and potyvirus-specific primers 353G1, 353G2 (Petrzik, K., unpublished data), P9502, P0502 (Revers et al., 1999) and Poty2/P4 (Gibbs and Mackenzie, 1997). Successful amplification with potexvirus-specific primers, sequencing and comparison with GenBank sequences revealed White clover mosaic virus (WClMV) there. Coat protein gene of this virus was obtained with amplification and compared with complete sequence of WClMV from GenBank. O-strain of WClMV was identified.
