Influence of the preparing of a biopolymer-surfactant system on its aggregation behavior.
Pilgrová, Tereza ; Sedláček, Petr (referee) ; Pekař, Miloslav (advisor)
To study the effect of the method of preparation of biopolymer-surfactant on the aggregation properties several experiments were designed. Closer investigation was made on native hyaluronan-cetyltrimethylammonium bromide (CTAB) system. The system has been studied in the phosphate buffer in aqueous and saline solutions. The effect of the order of substances addition in the samples, the influence of different methods of preparation of certain concentrations of the solutions or the effect of dissolving powdered biopolymer and surfactant on the contrary, the resulting system were investigated. Fluorescence intensity was measured for all prepared samples and its changes were investigated. Pyrene and Nile red were used as fluorescent probes. Some samples were also measured by tensiometer. Measurement of surface tension was based on the method of divorcing the ring from the surface. It was found that the order of the mixing of solutions has a significant effect on the aggregation properties of the system and its appearance. Further effects were detected on the prepared samples by dissolving powdered biopolymer surfactants and vice versa, and environmental influences of the polarity of the system. It was found that sodium chloride abolished the effects of interactions in the system. Effect of method of preparation of certain concentration of CTAB in phosphate buffer is reflected only slightly.
Soil salination and natural lignite as a potential remediating agent
Kořínková, Alena ; Sýkorová, Ivana (referee) ; Pekař, Miloslav (advisor)
This bachelor thesis deals with sorption ability of raw and water washed lignite for salt NaCl from water solution. Measurement was carried out in accordance with norm ČSN 11265 Quality of soil – Determination of electric conductivity with some modification (time period, extraction agent). Behaviour of lignite-water-NaCl, soil-water-NaCl and soil-lignite-water-NaCl mixtures was studied by measuring pH and electric conductivity.
Hyaluronan for targeted drug delivery
Ureš, Tomáš ; Vávrová, Milada (referee) ; Pekař, Miloslav (advisor)
Hyaluronic acid (hyaluronan, HA) is a linear polysaccharide formed from disacharide units containing N-acetyl-D-glucosamine and glucuronic acid. HA is present in almost all biological fluids and tissues. In many cancer cells there is an upregulation of CD44, a receptor that binds HA. The receptor CD44 collocates hyaluronan in the special way. The bioconjugates built-up from the cytotoxic substances and hyaluronan are called prodrugs. The prodrugs can be used for targeted drug delivery system. Their main advantage is there is no need to medicine so high doses of pharmaceuticals and their adverse side effects are minimized. Hyaluronan can be used as well as the intra-articulation injection by osteoarthritis. HA is also used as the nutrition of joints, by treatment for diverse dermatic lesions, the repair of scars and it can be the ingredient of eye drops and nasal sprays. The cosmetic manufactures add HA into anti-wrinkle creams and anti-aging care.
Flow properties of hyaluronan colloids
Gabrielová, Michaela ; Ptáček, Petr (referee) ; Pekař, Miloslav (advisor)
The purpose of this study is to zoom in implication of flow properties of hyaluronan colloids with respect to physiological role. In the first part I will introduce physical formulation and definition of flow properties and reology, process to its measurement and utilization. In the second part you can read about hyaluronic acid and colloids of its salts. In this part I will focus on first studies of hyaluronic acid throught its occurence to its chemical properties. Within this work I will try to demonstrate interconnection of both subjects.
Fluorescence quenching in colloid research
Srholcová, Barbora ; Ing. Michal Zavadil, Ph.D (referee) ; Pekař, Miloslav (advisor)
Allready known theoretical observations about utilization of the emission quenching of fluorescence probes in the study of colloids were summarized by finding in the literature search and on the internet, especially the determination of the probe´s location, the aggregation numer and finally the effective viscosity. In the experimental part was studied quenching of the pyrene in the medium of the anionogenic and non-ionic surfactants. As a quenchers were used iodobenzene, tetracyanoethylene and cetylpyridiniumchloride. Selected experiments helped by finding out the aggregation number of the micelle.
Hyaluronan nanofibers
Linhartová, Beáta ; Velebný, Vladimír (referee) ; Pekař, Miloslav (advisor)
This bachelor´s thesis deals with the most common methods of producing nanofibres, it presents an overview of these techniques and briefly desribes common models. It shows their advantages and disadvantages and tries to follow the develompent of nanofibres. This theoretical part is followed by an experimental one which describes concrete example of producing nanofibres.
Development and optimization of stable recipe of tanning lotion.
Němečková, Lenka ; Pávek, Stanislav (referee) ; Pekař, Miloslav (advisor)
This bachelor´s thesis is focused on problems of development and optimalization of stable formula of sunscreen with a sun protective factor SPF 15, which is produced in Altermed Olomouc company. In theoretical part are defined terms as cosmetics, laws, which deal with cosmetics and sunscreens especially. Somatic types of the skin, their reaction to UV rays and their protection against them are there described as well. In experimental part there summarized all requirements, which are necessary to realize for introduction of preparation into the market place and during long term use it must continue to pass all laws, health, marketing and functional claims. I produced a sample of stable sunscreen with protective factor SPF 15 in practical part. This sample was tested in accredited laboratory for all aspects which are necessary for legislation. The results of these testing are noticed in laboratory protocol.
Non-energy uses of lignite
Zedníčková, Petra ; Taraba, Boleslav (referee) ; Pekař, Miloslav (advisor)
This bachelor thesis is focused on the meaning of coal and it's utilize. It is especially concentrated on lignit and it's utilize out of energetic sector. Lignit constitudes geologicly youngest type of coal and belongs to caustobiolits , which is solid with various degree of coalification of original phytomasa. Degree of coalification is situated between peat and lignit. Lignit is characteristic by clearly well-kept structure of wood. Exactly this relative youth of lignit is reason for it's interesting qualitative indexes, in which belongs especially it's natural sorptive ability and quite high volume of humic acids. Both of these characteristics cohere together, because humic substances will certainly be main reason for sorptive characteristics. Coal, in comparing with other natural material, is by it's structure very complicated mixture of both organic and inorganic substances in various states of matter. It is matter of heterogenous system of macromolecules, which is structured of microscopicly distinguishable and chemically various substances. It can be composed of organic components or work as mineral admixture.
