Mechanisms coordinating auxin metabolism and transport
Novotná, Lenka ; Petrášek, Jan (advisor) ; Serre, Nelson Bernard Calixte (referee)
Auxin is a small molecule that functions as a plant hormone, and it exists in several forms, of which indole-3-acetic acid (IAA) is the most studied one. IAA modulates cell elongation, division, and differentiation by generating local gradients, and it is essential for almost every aspect of plant growth and development. These gradients are established by the cooperation of IAA biosynthesis, metabolism, and transport. A plant responds to both local auxin maxima and minima; thus, it is necessary to regulate auxin metabolism and transport tightly. However, lots of studies show the roles and regulation of auxin metabolism and transport separately, providing quite rarely discussion on the cooperation of these two processes. Hence, this thesis aims to sum up and refer to mechanisms and regulation of auxin metabolism and transport as a whole, rather than separately, and underline the importance of the cooperation of both auxin metabolism and transport in the plant development.
Study of human pepsin phosphorylation
Novotná, Lenka
Novotná L.: Abstract of Ph.D. Thesis Protein phosphorylation is one of the most important post-translational modifications in nature. The process of protein phosphorylation/dephosphorylation regulates almost all cellular processes and plays a key role in the maintaining of cell homeostasis. Abnormal phosphoryla- tion is associated with many serious human diseases including cancers. The phosphorylation of main gastric aspartic proteases pepsins and their precursors varies among animal species. Pepsins and pepsinogens of several species contain phosphate groups, while others have no phosphate groups. Phosphorylation of human pepsins and pepsinogens is probably associated with different gastric diseases. Gastric cancer patients show higher phos- phorylation degree of pepsinogens compared to healthy subjects. To utilize this finding for diagnostic purposes, it is requisite to elaborate simple and reliable methods for the study of pepsin and pepsinogen phosphorylation, which could be applied to limited sample amounts. This thesis focuses on the elaboration of appropriate methodical approach for the study of protein phosphorylation, particularly phosphorylation of human pepsins, which are characte- rized by a very low content of basic amino acid residues and a higher content of acidic amino acid residues.
S cílem zjistit vliv procesu byly v této práci porovnány vzorky připravené pomocí metod: freeze-casting, izostatické lisování za studena, uniaxiální lisování a suspenzní odlévání.
Impact of macro channels on mechanical stability of bone scaffolds during indirect 3D printing
Vojníková, Michaela ; Novotná, Lenka (referee) ; Salamon, David (advisor)
Porous materials are currently subject to the great interest of tissue engineering. They provide unique properties such as bioactivity, biodegradability, osteoconductivity, and vascularization. Particularly, ceramic porous systems show appreciable potential in medical applications. However, there is a crucial problem with the porous scaffold on account of their bad mechanic properties and therefore they are presently used only at low-load locations. This thesis focuses on the preparation of the scaffolds made of hydroxyapatite by using the freeze-casting method where the indirect 3D printing was applied to get open channels with the size over 200 µm. It also compares the mechanical properties of the scaffolds with different internal structures and monitors how the implementation of different types of grids affects the resulting stability. The scaffolds were prepared with a different arrangement of macro-channel in the internal structure, but they had equal dimensions as common property. The 3D grid was implemented before freezing into the mold and afterward the grid was eliminated by sintering, leaving only a channel system with the size 540-600 µm in the final scaffold. The influence of the type of the 3D grid on the resulting mechanical stability of the scaffold was determined. Rotation of this grid does not have a significant effect on the result, while it only helps with cracking in the direction of the helix. The combination of these methods reports very good controllability with directed macro-channels in the resulting scaffold and therefore it is suitable for the preparation of the bone-implants with different structures.

Šantavý, Tomáš ; Novotná, Lenka (referee) ; Salamon, David (advisor)
Bachelor thesis focuses on preparation of hydroxyapatite ceramics by freeze-casting. Hydroxyapatite is an important ceramic material, due to its biocompatibility, biodegradability and bioactivity it has a wide range of use in medicinal applications especially in replacement of bone material. The chosen method, freeze-casting, is flexible, eco-friendly and we are able to achieve a high level of porosity. The method includes freezing, freeze drying and sintering which creates a porous structure of the prepared material. Objective of this bachelor thesis was to find out the impact on the created porosity and mechanical properties of the final material. Freezing rates were applied 3, 4, 5, 6, 7 and 26 m/s. During the experimental part it was established that the freezing rate has a significant influence on the newly formed inner microstructure, it directly influences the interlamellar distances and mechanical property which is compressive strength. With liquid nitrogen with a speed of 26 m/s the achieved compressive strength was of 15 MPa as well as the shortest interlamellar distances of 14-22 µm. With slower freezing rate the average interlamellar distances were 40-150 µm and compressive strength of 2-4 MPa with comparable porosity.

A number of partial restructuring measures are offered to optimize business operation to improve its efficiency and performance. The structure inside the company is one of the most important tools for managing its performance and its correct definition, and a proper and meaningful distribution can play a key role in the competitive environment. In many cases, the requirement for optimization of operation will arise from the requirements of facility management, and therefore restructuring measures relating to organizational changes should be implemented with respect to the principles of facility management or should be based on its optimization. This thesis presents the impact of restructuring measures by centralizing of divisions on optimizing its operation with respect to the facility management. The aim of the dissertation is to evaluate the organizational change in the company with several divisions, taking into account the principles of facility management. The result of this thesis are the methodical recommendations during the establishment and evaluation of the division´s centralization in the company. It is based on the findings from the literature review and the real change in the building company.
