Characterization of biological and functional features of a new type of CD27- memory B lymphocytes.

The increased frequencies of two novel B cell populations defined as IgM+ CD19+ CD27- CD21low CD38low CD24+ and IgM+ CD19+ CD27- CD21low CD38low CD24- in peripheral blood of patients with common variable immunodeficiency (CVID) compared to healthy donors were found. The aim was to search for such B cells in patients with rheumatoid arthritis (RA) and their further characterization. The production of immunoglobulin (Ig) mRNA in single B cells was analyzed using flow cytometry, single cell sorting and RT-PCR, IgVH-specific PCR, cycle sequencing and statistical analysis. The study was focused on analysis of variable regions of the heavy chains of Igs and significant differences in the usage of VH, DH and JH gene segments, mutational frequencies, distribution of silent and replacement mutations, length and composition of CDR3 regions, clonal relation and RAG gene expression in above mentioned B cell populations were found. Because of lack of the surface CD27 molecule being regarded as marker of B cells that have undergone antigen-driven germinal reactions, analyzed populations were considered as naive. Genetic, humoral and cellular factors of development of autoimmune diseases

Autoimmune diseases currently represent the most serious medical issues, mainly due to generally increasing number of patients with these diseases. Their pathogenesis is likely caused by hereditary factors, cellular and humoral interactions influenced by external environmental factors, whose knowledge is important both for diagnostics and therapy, and for theoretical immunology. The aim was to examine the correlation between genetic alterations and the production of autoantibodies, clinical manifestation of the disease; to identify joint autoantigens, further to demonstrate development of leukemic cells from originally autoreactive B cells, the role of B lymphocytes in the pathogenesis of the disease and finally to develop a method for detection of B cells recognizing a defined autoantigen. Several predisposing polymorphisms were revealed using genetic analysis however, they were not exclusively associated either to clinical forms of the disease or usable as prediction markers. This bachelor work pursues the hidden advertisment and mainly Product Placement. The work will make us acquainted with terms of hidden advertisment, product placement and it's positioning in the marketing communication. Theoretical part targets on definition of terminology like advertisement, hiden advertisement, product placement and legal regulations. The work deal with the analyses, the impact of the legalization of product placement in Czech media after receiving the direction of EP and Counsel 2007/65/ES and at the end it assess whether Product placement to ban or to allow in television programmes.
