Mechanismus působení polymerních léčiv na bázi HPMA

However cytostatic polymeric conjugates based on N-(2-hydroxypropyl)methacrylamide are in clinical trials, we are still far away from understanding the mechanism of their action. Cleavability of the drug from oligopeptidic side-chains of polymeric carriers seems not to be necessary condition for pharmacological activity and those spacers play most probably a very important role in the accumulation of conjugates in tumor cells.

Conjugates of N-(2-hydroxypropyl)methacrylamide with doxorubicin containing human IVIg were used for palliative treatment in patients with advanced cancer. More thena 116 biochemical and immunological parameters were tested. A decrease of some parameters with patological values was seen, and physiological values were occasionally reached, including some TAA.

Water-soluble synthetic polymers and their conjugates with antibodies, oligopeptides or saccharides proved a potential targetable drug delivery system. Recently, copolymers of N-(2-hydroxypropyl)methacrylamide (HPMA) have been developed in our laboratory as polymeric carriers of drugs.
