Steganalysis is the opposite science discipline of the steganography - which is an art of information hiding . Steganography deals with embedding of secret messages to different types of media. The most commonly used cover media in modern steganography are image files . This bachelor's thesis creates an overview of known steganograpy and steganalysis methods . It also describes practical implementation of application that is able to detect hidden information in image.
Epignetic Modifications of the Sperm and the Application in Clinical Practice of Human Assisted Reproduction Therapy

Basement of healthy embryo development comes from quality of oocytes and spermatozoa. Today, when percentage of couples suffering infertility together with assisted reproductive therapy (ART) is increasing, understanding to gamete biology and heritable epigenetic code is crucial. The study is focused on promising epigenome based markers that could serve as indicators of gamete quality for either their screening or selection for ART. Accordingly selected markers were used for the investigation of environmental pollutant bisphenol S (BPS) effect on gametes quality. To obtain these aims, we have used human semen samples, boar semen samples and ICR mice gametes. Samples were analyzed by flow cytometry, immunocytochemistry and western blot analysis. All experimental work was in accordance with Ethics committee University Hospital in Pilsen and approved experimental designs for appropriate experimental animal project. In the study, we detected the dimethylation of histone H3 on lysine K4 (H3K4me2) as potential epigenetic marker of sperm quality and chromatin immaturity. The Role of Cellular Metabolism in Carcinogenesis. Molecular Pathophysiology of Bladder Cancer Chemoresistance

Therapeutic resistance of tumours represents an important clinical issue. We can classify the therapeutic tumour resistance in two ways. According to the clinical course, tumours can behave either as primary resistant, i.e. from the very beginning not responsive, or they can display a secondary (also called acquired) resistance, whereby an initial clinical response is lost and the tumour develops into chemo-, radio- or immunoresistant disease. An alternative classification distinguishes cell autonomous resistance mechanisms from resistance that relies on complex interactions within the context of tumour microenvironment. From the research perspective, modelling therapeutic resistance frequently involves experimental treatment of sensitive cancer cells and selection of daughter resistant cell lines. The Ph.D. thesis includes derivation of two unique models of urothelial bladder carcinoma therapeutic resistance. The first model involves newly established urothelial carcinoma cell lines BC44 and BC44DoxoR, which resulted from a prolonged doxorubicin exposure of the mother cell line. The role of SIRT1 during in vitro maturation of oocytes

SIRT1 histone deacetylase acts towards many epigenetic and non-epigenetic targets. The involvement of SIRT1 in oocyte maturation is assumed and the importance of ooplasmic SIRT1 pool for further destiny of matured oocyte is strongly suggested. We hypothesized that SIRT1 play role of the signal molecule in mature oocyte through selected epigenetic and non- epigenetic regulation. We observed SIRT1 re-localization in mature oocyte and the association with spindle microtubules. In matured oocyte, SIRT1 shows a spindle-like pattern and spindle- specific SIRT1 action is supported decreasing α-tubulin acetylation. Based on the observation of histone code in immature and matured oocytes, we suggest that SIRT1 is mostly predestined for epigenetic mode of action in germinal vesicle (GV) of immature oocyte. Accordingly, SIRT1- driven trimethylation of histone H3 on lysine K9 in matured oocyte is considered to be an inheritance of GV epigenetic transformation. Taken together, our observations point out the dual spatiotemporal SIRT1 action in oocyte capable to be switched from epigenetic to the non- epigenetic mode of action readily depending on meiosis progress. Digital Text Steganography

The goal of this bachelor project was to learn about digital text steganography, about methods for hiding the text, to implement my own or already existing methods and finally to compare acomplished results of these methods. This project introduces four methods (Zero distribution, semathics, syntax and Format based) which hide secret data into cover text, each of them by its own way. Research has found that semanthics method produces the best properties of stego text followed by Format based and Zero distribution, syntax method ended up worst. Based on the data found we can choose which method we will use.

DNA damage response in mammalian oocytes

During early embryonic development oocytes are arrested in prophase I of the first meiotic division, in which they can persist for years. After reaching sexual maturity and the luteinizing hormon surge resumption of meiosis and meiotic maturation occur. Oocytes are arrested again at metaphase of the second meiotic division. At this stage they are ovulated and waiting for a fertilisation. Oocytes are during their development exposed to factors that cause DNA damage, of which DNA double-strand breaks (DSBs) are the most serious threat. The maintaining of genome integrity is crucial for quality of oocytes, fertility and proper embryonic development. The mechanism of the oocyte response to DSBs presence is not fully understood and it seems to differ from somatic cells. We assume that DSBs are repaired during meiotic maturation probably by a mechanism of homologous recombination (HR). In this thesis we focuse on essencial recombinase RAD51, which participates in the repair by HR. We found that RAD51 inhibition leads to an increase of segregation errors in anaphase I. Using high resolution live cell imaging we observed chromosomal fragments and anaphase bridges. Immunofluorescence detection of DSBs-marker γH2AX showed increased amount of DSBs in prophase I and MII stage after RAD51 inhibition. Our data...
Technical Condition Monitoring of Radar Systems Components

The goal of this thesis is to design a diagnostic subsystem for secondary surveillance radar and to construct a functional prototype of communication and testing generator unit. The first part of the thesis aims at description and division of radar systems, with emphasis on air traffic control radars.  Main attention is being focused on ATC secondary surveillance radars and description of their function. Following part deals with requirements specification for design of diagnostic system and the system design itself. Part of the thesis forms design, production and testing of prototype of block enabling communication of diagnostic subsystem and function of testing generator for secondary surveillance radar.

Universal Sensoric Platform with Programmable Features

The aim of this thesis is to design universal sensoric platform for data collection in manufacturing environment. Device is capable of collecting data from variety of sensors, but it is also able to control external systems. Platform is controled by Lua scripts. The scripts are generated in user friendly visual programming environment Blockly. Platform is based on WiFi module ESP-WROOM-02.
