A Potential Neuroprotective Action of Ghrelin on Dopaminergic Neurons Transplanted into the 6-hydroxydopamine Lesioned Rat

Parkinson's disease is a neurodegenerative chronic disorder which is characterised by loss of dopamine in nigrostriatal pathway in the brain. One of the alternative therapies is transplantation of embryonic ventral mesencephalon tissue to the affected brain area but currently grafted cells do not survive the transplantation process very well. Ghrelin, which has neuroprotective properties could be a potential additional treatment after transplantation with the intention of increasing the number of live cells in the graft. The aim of this study was to elucidate a presence of ghrelin receptor in embryonic ventral mesencephalon used in transplantation and assess behavior of rats after transplantation. Immunocytochemistry was performed for ghrelin receptor, ghrelin-O-acetyl transferase, tyrosine hydroxylase and for neural and stem cell markers in embryonic ventral mesencephalon suspension. 50 Sprague-Dawley rats with 6-hydroxy- dopamine lesions were split into five groups; Group A was non-transplanted group, other groups underwent transplantation of embryonic ventral mesencephalon and received diverse treatments (ghrelin agonist, low doses of ghrelin, high doses of ghrelin or saline1 ). Depression, antidepressants and diabetes mellitus - does exist mutual relation?

This bachelor's thesis first briefly describes the relationship to the quality of life of diabetes. The main part deals with the mutual relationship between depression and diabetes including a basic pathophysiological mechanisms. A selection of clinical trials on the problem solving is followed. Finally, it provides a brief overview of the antidepressants in patients with diabetes and their side effects with special attention to the influence of body weight. Diagnostics of the enzymatic defects in patients with coeliac disease

Celiac sprue is a chronic, lifelong disease characterized by permanent intolerance to gluten. Small intestinal mucosa of these patients shows typical inflammatory changes. These changes lead to reduced absorption of nutrients, minerals and vitamins that may result malabsorption syndrome. Diagnosis of celiac disease must be a comprehensive evaluation of the results of immunological testing of autoantibodies and microscopic evaluation of the intestinal mucosa state. The bachelor thesis deals with the histochemical examination of intestinal biopsies, which allows better distinguish malabsorption syndromes of different origin. The aim of the study was to determine incidence of celiac sprue and evaluate the completeness of remission in patients on a gluten - free diet in patients aged over 50 years in intestinal biopsies samples taken in 2013. Strict gluten - free diet leads to reparation of mucosa and brush border enzyme activity. Biopsies were snap frozen immediately after collection, to preserve the morphology of tissue and the brush border enzyme activity. Reactions, demonstrating the enzymatic activity of the lactase, trehalase and dipeptidylpeptidase IV, were exercided on sections by previously described histochemical methods. These proteins are involved in the repairs of errors that happen during the transcription of genes. Inheretted mistake of any of these genes clinicaly displays usually in the the form of so-called Lynch syndrom. The most typical presentation at the molecular-genetic level is microsatelite instability of tumor cells. In the theoretical part Lynch syndrome is described, etiology, forms of Lynch syndrome, diagnosis and prevention possibilities. In the practical part a test of the group of 30 patients with positive colon cancer. I describe a preparation of immunohistochemical slides and study results.

A survey of anatomy and physiology of the basal gaglia is also included. The main part gives a special attention to Tourette's syndrome. Brjančinová Lenka Searching multiple pregnancy using biochemical markers in the early stages of pregnancy Bachelor theses Charles university in Prague, Faculty of Pharmacy field of study: medical laboratory technician The bachelor's thesis objective is to work out an algorithm enabling multiple pregnancy detection by second-trimester biochemical markers of alpha-1-fetoprotein and human chorionic gonadotropin in the blood serum of women pregnant with one foetus or twins. Detected markers' values are converted into MoM values, adjusted to body masses of expectant mothers and subsequently entered into graphs where shifts in values between single and twin pregnancies are visible. In testing several empirical methods, the algorithm was found by which 78,75 % multiple pregnancies can be proved in the studied group, with false positive ratio of 2,50%. The effect of mud-bath applications on recovery from experimentally induced inflammatory lesion

Fieldof study: Medical laboratory technician, combined study from Aim of the thesis: The aim of the experiment was to clarify the effect of mud baths on experimentally induced subacute inflammation of the right hind paw of rats of both sexes. In the experiment, we tried to distinguish the influence of therapeutic mud and the effect of heat itself. Material and methods: We mixed 5 g of paraffin oil with 5 g of lanolin and 5 ml of heat-killed Streptococcus pyogenes B stock. The suspension is homogenized for 60 minutes in an ultrasonic bath. Induction of arthritis: Arthritis was induced by two subcutaneous injection 100 μl of the prepared suspension applied to the right hind paw (spaced by 8 days). Experiments were conducted on adult Wistar rats. The rats were divided into 5 groups of 6 male and 6 female. During the experiment were 4 to 5 times a week for 20 minutes (total 34x) placed in rearing containers with different materials. Conclusion: Differences were observed between the sexes in the healing of inflammation in different baths. Weight reduction is almost always an important measure when treating obese diabetics, since it contributes to reducing the complications related to obesity and diabetes. Objective: The objective of the thesis was to evaluate the results of weight reduction in morbidly obese patients with diabetes mellitus type 2 achieved by a combination of low-carbohydrate diet and mediterranean diet. Furthermore we assessed diet questionnaires with the goal of finding and correcting mistakes in eating and exercise habits. Title of Thesis Signs of epilepsy spectrum disorder in a selected population sample The thesis deals with comparing occurence of signs of epilepsy in a population sample of healthy people and people with a specific disorder (smokers, depression, bipolar disorder, headache). A questionnaire was chosen as a research method. It was proven that the occurrence of an abnormal score is higher in the depressive population. The results in a population with headaches and a bipolar disorder are inconsistent. A higher occurrence of an abnormal score was not proven in the population of smokers. The temporo-limbic dysfunction is a very broad issue, the results have to be verified by another researches. The goal of the thesis was to compare results of the two researched groups - healthy people and people with a specific disorder.
