The effect of 6S-like RNAs on physiological differentiation of Streptomyces coelicolor

The variety of bacteria and their genomes sometimes causes conservation of homologue molecules to be displayed not in sequence but in secondary and tertiary structures. In the case of the regulatory 6S RNA, sequence homologues have been found in over 100 bacterial species so far. However, none were found in the genus Streptomyces. The unique genome of these soil- dwelling bacteria, known for their capacity to produce antibiotics, has a high G/C content and diverges substantially from distantly related bacteria. Yet in the non-coding 6S RNA it is the secondary structure that is crucial for its function. The 6S RNAs trap sigma factors by mimicking target promoter sequences in order to help with switching sets of expressed genes during developmental transitions. 6S-like RNA genes in Streptomyces coelicolor have been computationally predicted by comparison of in silico modelled secondary structures of known 6S RNAs. The aim of this thesis was the verification of these 6S-like RNA predictions. The experimental approach was based on RNA co-immunoprecipitation (RNA CoIP), as well as RT- PCR from RNA samples. Preparation and testing of SNOM probes

The area of research that deals with surface modification and preparation of nanostructures is still very unexplored. And only a little contribution to this field is discussed in this bachelor thesis. Its goal is to manufacture and to test a probe made off hollow optical fibre that is used in Scanning Probe Microscopy. Optical fibre parameters in combination with proper and unique techniques allow the breakthrough off very interesting applications. It's worth mentioning the gas injection in the proximity of the surface of a sample (GIS) and thus its modification by use of electrons (FEBID), ions (FIBID) or laser beams.
Multimedia module for radio controlled models

The thesis deals with the development and realization of a multimedia module with a microcontroller for radio controlled models of ships, cars, and fighting techniques. The module uses commands from the RC transmitter to control the model illumination and accompanying sounds. The thesis summarizes principles generally used in RC models. Based on these general principles, a new control element for the RC transmitter is designed. A relevant multimedia module is designed and implemented. The last part of the thesis is devoted to the description of the basic parts of microcontroller firmware.
Heating of administrative building

The dissertation deals with heating of the administrative building. The building is heated with the floor heating and the panel heating units. The heating source is an air to water heat pump with a bivalent heating from the central heat supplying (CZT). I deal in the project with design, dimension and connection of all these parts.
Profile-Guided Optimizations for Instruction Caches

Instruction cache performance is very important for the overall performance of a computer. The placement of code blocks in memory can significantly affect the cache miss rate. This means that a compiler can improve the performance of a program by placing parts of code at the right addresses in memory. This work discusses several methods for collecting profile information, and describes an algorithm that uses profile information to guide code block placement. Additionally, the algorithm is added into the optimizer of the LLVM compiler, and improvements in cache performance are evaluated.
Design and realisation of solder-ball mounting device

This work deals with issues of design and construction of facilities for mounting solder balls. The first part includes a familiarization with the types, manufacture and assembly of solder balls in practice. The following section includes the actual construction work on the equipment, procedures, considerations and finish work on the plant. The last part provides information about solder balls testing, which are mounted on the FRITSCH station, and compares potential errors.

The functional trans-translation system is important to allow survival of antibiotic producing microorganism under unfavourable conditions. The ssrA genes from S. coelicolor, S. collinus, S. aureofaciens and S. griseus were amplified and subjected to sequence analysis. To investigate the role of tmRNA in streptomycetes we have examined the level of tmRNA during the growth and differentiation under various physiological conditions
Aktivace translačního systému v průběhu buněčné diferenciace streptomycet

Synchronously germinating spores of S. granaticolor were used to study the activation of translational system and expression of proteins. Proteomic approach was used to gain more information on expression of proteins during germination on two types of media differing in ability of cells to produce antibiotic granaticin. The requirement of functional trans-translational system for reactivation of translation in S. aureofaciens exposed to unfavourable conditions was examined. Abundance of tmRNA is connected with changes in the rate of translation and with the accumulation of tetracycline. Our data indicate that trans-translation could be involved in self-defence against antibiotics synthesized

Práce se zabývá zajišťovacím účetnictvím jednak v rovině teoretické, jednak v rovině praktické. Teoretická část vysvětluje základní principy zajišťovacího účetnictví v souladu s Mezinárodními účetními standardy IAS 32, IAS 39 a IFRS 4. Praktická část je zaměřena na řešení problematiky vykazování zajišťovacích nástrojů v podmínkách bankovního sektoru.
