Polymorphism of the transcription factor NF-κB and Toll-like receptor 2 in a production population of cattle (Bos taurus L.)
Samaké, Kalifa ; Novák, Karel (advisor) ; Vinkler, Michal (referee)
The broader purpose of the work is to find and interpret polymorphism in the genes of natural immunity of cattle to be used to improve disease resistance. The NGS method on the PacBio platform was applied for the resequencing of the gene for the key receptor of innate immunity TLR2 and two genes coding for the components of the downstream transcriptional factor NF-κB. In the population of 149 bulls of the Czech Simmental breed, 22 polymorphisms were found in the gene NFKB1 (5 new), while in the NFKB2 gene 13 SNP were found (10 new). 21 SNP were found in the TLR2 gene (3 new). Of the 56 found polymorphisms, 6 SNPs were nonsynonymous. One SNP leads to a change R474G in the NFKB1 product and five to changes E63D, R152Q, I211V, R563H and H665Q in the protein TLR2. Knowledge of the haplotypes facilitated the development of individual genotyping reactions. In TLR2, a high number of haplotypes was detected, both from the PacBio reads and the statistical reconstruction. In addition, two clusters of haplotypes were ditinguished inTLR2, possibly due to diversifying selection or introgression. The knowledge of genetic diversity in the population allows for the planned association studies with health data. Localization in functional domains allow to define the change with the greatest effect, in particular...
Archaeological evidence of medieval armour in the Czech Republik
Interrogation tactics
Novák, Karel ; Konrád, Zdeněk (advisor) ; Musil, Jan (referee)
INTERROGATION TACTICS The purpose of this diploma thesis is to systematically summarize knowledge and information about fundametal tactics and methods of an interrogation used in interrogation practice. Knowledge of these procedures is necessary for proper execution of an interrogation and also is suitable for legal practice. The thesis consists of 4 main chapters, each of them deals with relevant issues within the field of an interrogation. The First chapter "Interrogation as a forensic method" includes general discussion about an interrogation and a statement such as definitions of basic concepts, definition of a nature and a purpose of an interrogation. In addition to further this chapter also contains analysis and evaluation of Czech legislation governing the interrogation. The Second chapter "Preparation of an interrogation" is focused on a preparation of an examiner before an interrogation as one of important elements to succesfully realize itself interrogation and to achieve its goals. The Third Chapter "Interrogation tactics" first concetrates on particular phases of an interrogation namely an introduction phase, a monologue and a dialogue where emphasize issues of questioning in practice. Attention is also paid to various situations that may occur during interrogations. This part of third...
Electrophysiological Models of Seizure like Activity for NO Detection in Vitro
Novák, Karel ; Konopková, Renata (referee) ; Otáhal, Jakub (advisor)
Definition of work: Electrophysiological models of seizure like activity for NO detection in vitro Supervisor: MUDr. Jakub Otáhal, PhD. Department of Anatomy and Biomechanics FTVS CUNI Department of Developmental Epileptology CAS, Institute of Physiology Consultant: Mgr. Renata Konopková Department of Developmental Epileptology CAS, Institute of Physiology Aim: The aim of the thesis is to introduce nitric oxide (NO) and its role in physiology and pathophysiolgy of central nervous system (CNS), with the intention of epileptiform activity in the nervous tissue. Our research was realized on rat hippocampal slices in vitro and it compares the experimental biochemical models (low-Mg2+ , high-K+ , Carbachol, Bicuculline and 4-Aminopyridine model). These changes in the environment of slice should evoke the epileptiform activity in the nervous tissue of the rat. The final aim of the thesis is to create an experimental model of epileptiform activity in vitro in our setup for imaging techniques and NO detection. Methods: Experiments were performed in rat hippocampal slices in vitro. Rats were deeply anaesthetized with ether and decapitated. Brain was rapidly removed from the scull, cut in oxygenated (95%) and cooled (0, 5 řC) artificial cerebrospinal fluid (ACSF) using vibratome in coronal slices of 400µm of...

Public Support for Applied Research and Development from the Perspective of selected Business Entity
Novák, Karel ; Musil, Martin (advisor) ; Krbová, Jana (referee)
This thesis analyses the causes of states interventions in the market environment and the risk associated with them, in particular with the state support of research and development. There are several types of support, each of them has benefits on the one hand and inducted costs on the other one. The thesis assesses the benefits and inducted cost of the various types of state support of research and development from the perspective of the company Vyzkumny ustav organickych syntez. In the case of this business entity there is the economic analysis performed and the decision rule for the most appropriate aid derived. At the same time thesis tries to find how to increase efficiency of the government support.
Science and Research as an Important Positive Externality
Novák, Karel ; Musil, Martin (advisor) ; Krbová, Jana (referee)
The work focuses on the issue of research and development funding from public resources, mainly on the project funding of applied research. The aim of the work is to describe some aspects, which were brought to this area by Science, research and innovations reform approved by the Czech Government on 26th March 2008. Public intervention in this area is discussed in terms of the basic economic theories. The efficiency of public funds spending is discussed in terms of the cost-output-quality output-benefit chain. The part of the work is a comparison of the state funding of the applied research and development in the Czech Republic and other countries. The work tries to find some connections between changes in the way of assessing research quality and direction of financial flows in this area.
