Structure and properties of bacterial biofilms

This thesis deals with the study of basic properties of cells of bacterial biofilm produced by PHA producing bacteria, specifically Burkholderia cepacia and Burkholderia sacchari. Two cultivation approaches were used to compare different types of cells: at first, the cells were cultivated in bioreactor providing both planktonic and biofilm attached cells, secondly, cells grown on Petri dishes were also included in the study. In experimental part the affinity of each type of cells to hydrophobic phase was studied using BATH method. It was found that sessile cells, either biofilm associated cells as well as cells taken from Petri dishes, prefer hydrophobic phases more than planktonic cells. Also size and Zeta potential were measured using Dynamic Light Scattering (DLS). It was found for studied samples that, cells in these samples tend to aggregate and the difference of size between both types of cells occurs only for biofilm of the strain Burkholderia cepacia. Finally the impact of presence various bacterial cells types on surface tension of water as well as amount of PHA cumulated in different types of cells were observed.
The effect of magnetic field and other selected stressors on physiology of bacterial cells

This thesis deals with the effect of magnetic field and organic substances, namely benzene and p-nitrophenol, on cell of PHA producing bacteria Cupriavidus necator H16 and mutant strain Cupriavidus necator PHB4, which does not produce polyhydroxyalkanoates. Static magnetic field was generated by both permanent magnet and electromagnet. The effect of magnetic field on the growth of bacterial cells was studied using growth curves. It was found that cultivation in magnetic field and mineral medium mostly inhibits bacterial growth. Also the amount of polyhydroxyalkanoates was observed using FT-IR, flow cytometry and microscopy with fluorescent dye. Growth curves and flow cytometry were also used to study the influence of organic substances on bacterial cells. It was found that while benzene does not affect either C. necator H16 or C. necator PHB4, p-nitrophenol acts as the inhibitor of bacterial growth for both cultures. Finally the impact of p-nitrophenol on the accumulation of PHA was studied using gas chromatography.

The first part is theoretical, the second part describes the company's business and evaluates its valuation in the company. The third part contains proposals for solutions and the last part contains the conditions of implementation and benefits. Based on the performed analysis, the most suitable option will be chosen, which will be the best for the company.
Perception training in tennis

Title: Perception training in tennis Objectives: The main goal of this paper is to assess the impact of perceptual-cognitive training on the development of anticipatory skills in expert and novice tennis players. Methods: The result of this paper was obtained using an experiment based on the test-retest method. It is a randomized controlled trial in which the intervention groups underwent three weeks of perceptual-cognitive training in addition to the test and retest process. All parts of the experiment (test, perceptual training, retest) were based on watching edited video recordings of tennis serves using the temporal occlusion technique. The tracked variable in the experiment was the relative accuracy of responses. Results: In the intervention group of expert tennis players, a significant difference was reported between test and retest performance (p < 0.01; d = 1.05). No statistical significance was observed in the intervention group of tennis novices or in any of the control groups. Since the intervention and control groups of expert tennis players did not differ in the test stage, the observed increase in performance was attributed to the effect of applied perceptual-cognitive training. Mělník as the residence of Czech queens

The topic of this bachelor thesis is Mělník, a famous royal dowry town that used to be an essential residence of numerous queens of Kingdom of Bohemia. The focus is on the importance of Mělník throughout history, from its creation in the 9th century to the 15th century. The first two rulers to reside in Mělník were Przemysl princesses Ludmila and Emma. The last queen to ever stay in Mělník was Joanna of Rosenthal in the 15th century. This thesis is dedicated mainly to queens Elizabeth of Bohemia, Barbara of Cilli and Joanna of Rosenthal. The aim is to analyze available historical documents and secondary literature sources and use them to explain the connections of the queens to the town, portray Mělník as their residency and explain its economic and political effects. The use of art therapy practice for individuals with inadequate self-concept

This bachelor thesis deals with the use of art therapy in working with adults with inadequate self-concept. The thesis is divided into theoretical and practical part. The theoretical part contains two main chapters that deal with self-concept and art therapy. Self-concept deals with a person's relationship to himself, witch fundamentally influences the possibilities of how an individual can relate to the world and suceed in it. Art therapy in a narrower sense uses fine art for therapeutic purposes. The practical part reports on art therapy work with adults with inadequate self-concept. Research outputs are presented in the form of case studies.
Integrated information system of administrative activities

Tématem práce je charakteristika správních evidencí vedených v informačních systémech Ministerstva vnitra a dalších správních činností na úseku matriky a přestupků. Správní činnosti budou analyzovány z hlediska příslušné legislativy, která bude následně zhodnocena a bude navržena nová právní úprava. Cílem práce je ucelená informace o správních činnostech, analýza správních evidencí, ostatních správních činností a podání informace o poskytovaných údajích z informačního systému těchto evidencí. Práce se bude dále věnovat informacím o cestovních dokladech s biometrickými prvky a přípravě projektu. Na základě provedeného výzkumu bude podána prezentace výsledků analýzy získávání informací o správních evidencích a ostatních právních činnostech. V této části se práce bude dále věnovat stupni znalostí správních evidencí a ostatních správních činnosti, zdrojům informací o správních činnostech, hodnocení poskytovaných informačních služeb, frekvenci využívání informačních služeb, obecné informovanosti o zákonech a vyhláškách správních činností a znalosti zákona o svobodném přístupu k informacím a to z pohledu občana. V závěru práce jsou uvedeny výsledky vlastní analýzy a jejich zhodnocení.
Development of tennis rules with an emphasis on serving

Title: Development of tennis rules with an emphasis on serving Objectives: The main goal of this paper is to assess the possible impact of removing the rule of the second service from professional tennis matches on their length. It also focuses on this change's feasible influence on the attractiveness for viewers. Methods: The result of this paper was obtained using indirect observation of video recordings from Australian Open 2021, Roland Garros 2021, and Wimbledon 2021. In addition, was used notation analysis of tennis matches from listed tournaments. For tracked variables were used the rally length after both the first and the second serve. Another variable was the time gap between the first and the second serve. Results: Acquired results of this work show that tennis matches would not be significantly shortened after the removal of the rule of the second serve in relation to other time aspects of tennis matches. This outcome serves as an argumentative foundation for expert discussion about the possible introduction of a change to the existing rules concerning the second serve in the game of tennis.
