Gene sequencing in patients with cancer anamnesis

Cancer is the second most common cause of death in the Czech Republic, of which 5-10% are occupied by hereditary cancer syndromes. They are caused by a congenital - hereditary mutation in one of the alleles of the genes, when after the second random intervention in the other allele hereditary cancer develops. It is important to distinguish between hereditary and sporadic carcinomas due to the high risk of inheritance of mutated alleles in the family. The indication may be, for example, the onset of the disease at a young age or the recurrence of the cancer in the family In my work I focused on the analysis and evaluation of data obtained by Sanger sequencing. The aim was to find mutations in the genes mentioned below and to evaluate their pathogenicity by comparison with databases. In the theoretical part of the bachelor thesis I deal with cancer in general and hereditary cancer. I specify the hereditary breast and ovarian cancer syndrome, including genes, that may cause this syndrome - BRCA1, BRCA2, TP53, PTEN, ATM, PALB2, I also deal with Lynch syndrome and the MMR gene system. Last but not least, I describe a familial adenomatous polyposis associated with the APC gene. In the research part I focused on the examination of selected areas of 18 anonymized samples in the gene PALB2 - exon 13 and in the gene BRCA2 - exon 10/4 and exon 11/12. Using the PCR method, I prepared the samples for Sanger sequencing, which then took place in GenSeq s.r.o. In the last part of my work I deal with the analysis and evaluation of the results using the BioEdit program and the NCBI database. I found a mutation in 5 samples - in 4 it was a deletion of one nucleotide with a conflicting interpretation of pathogenicity, the last mutation was pathogenic - causes hereditary breast and ovarian cancer syndrome, it was a nucleotide duplication.
Influence of body size and temperature on cardiac function in vertebrates

Heart, as main driving unit of circulatory system of vertebrates, ensure a great number of body functions by constant circulation of blood in every individual. This body functions are provided by transposport of energy in various forms and by transport of chemical substances. Number of these body functions are absolutely necessary in each momemnt of the lifetime. If we describe metabolism as speed of all reactions in body, then circulatory system is one of the key means to reach it. Metabolic rate increases with two basic factors, body mass and body temperature. It's not proportional relationship, but allometric increase is conditioned by power law. In addition, this exponential relationship is limited, in case of body temperature, by temperature range specific for each living organism. It would be expected that changes in size of body and body temperature might have strong influence to quality and function of circulatory systém and heart itself. However, metabolism is significantly influenced by some another factors like quality of adjacent surrounding. Organisms belonging to vertebrates habitated great number of various enviroments and they developed lots of adaptations to survive there. The distribution of medicaments by the nursing sister in the social wealfare institutes

This bachelor's thesis focuses on the distribution of medication in the social welfare health institution. The objectives of the thesis were to focus on procedures of distributing medication by nurses and to introduce the most frequent complications when distributing the medication in the social care institute. The thesis is composed of two parts, theoretical and practical. The theoretical part describes social and health care services, a health and social care system, a general pharmacology, medicaments' distribution and the complications during the distribution, and the competency of nurses. The research part concentrates on individual study and describes a methodology of research and its organization. This research was done via survey, and the results are analyzed and discussed. The Degree of Time Expressions Accuracy in the Texts of Non-native speakers of Czech in the learner corpus Merlin

1 Abstract The diploma Thesis will focus on the general description of the expression of temporal quantification by non-native speakers of Czech in written speeches. Based on professional literature, the graduate will define time indicators, in the next step she will analyze the most frequently used textbook series of Czech for foreigners - Czech step by step by L. Holá, in which she will monitor the presentation of time data, both grammatically and lexically in terms of the range of indicators with regard to the degree of language acquisition, but also in terms of the way of presentation of the indicators. In the next part, an analysis of the occurrence of time indicators in the Merlin student corpus will be performed. The scope of use of time indicators will be monitored with regard to the level of knowledge according to SERR, the degree of accuracy of the indication of time quantification and the error typology of time means.
The study of bryophyte species richness and community composition on a very fine spatial scale connected to in situ measured microclimate

The life of bryophytes is closely connected with their surrounding environment. Changes in atmospheric conditions on the microscale directly affect the physiological functions of bryophytes, which in turn determine their distribution. The current development of technologies allows us to measure the microclimate affecting bryophytes directly in the field. Nevertheless, there have not been many studies published examining the response of bryophytes to in situ measured microclimate. This diploma thesis is one of the first in Europe to provide data from continuous field microclimatic measurement performed on a scale relevant to bryophytes. It deals with the influence of the microclimate on the bryophyte species richness and community composition within a single gorge in the Bohemian Switzerland National Park. For 17 months, 38 HOBO Pro v2 Onset dataloggers were recording air temperature and air humidity 10 cm above the ground. I performed detailed bryological survey on two differently sized plots (circle with a radius of 1 or 2 m) around each of the dataloggers.
