Health effects of Czech beer

The aim of this bachelor thesis was to study physiological effect of various kinds of lager beer and to compare these effects in Czech beer and several beers produced in foreign countries. In theoretical part Czech beer characteristics and biologically active compounds in beer are described. Quantification of total phenolics, total flavonoids, antioxidant activity and bitter substances was performed photometrically. The quantity of vitamin C was determined by high performance liquid chromatography with spectrophotometrical detection. Beer buffering capacity was analyzed by pH measurement in artificial stomach and intestinal liquid, respectively. Analysis of physiological effect was accomplished by analysis of proteolytic enzyme activity. The total of 22 samples (16 Czech and 6 foreign lager beers) was studied. In these samples levels of basic technological characteristics and buffering capacity were compared. Alcoholic and non-alcoholic beers were compared too. According to experimental data it was confirmed that consumption of Czech beer (mainly lager beer) positively influences digestion process and activity of digestive proteolytic enzymes.
PREPARATION AND APPLICATION OF SOME BIOPOLYMERS, NANOPARTICLES AND NANOFIBRES FOR COSMETICS AND FOOD

The presented doctoral thesis is focused on preparation of nanoparticles and nanofibers with natural active ingredient and testing their biological effects. Modern types of application forms were prepared from biomaterials based on one or more natural polymers. Chitosan particles were prepared from cross-linked polymer using ultrasonication. A mixture of soy lecithin and cholesterol was used for preparation of liposomes. Poly-3-hydroxybutyrate was used for preparation of combined liposomes, too. All liposome particles were prepared by ultrasonication. Nanofibers were obtained from polyhydroxybutyrate using electrospinning. Mixtures of low-molecular antioxidants obtained by extraction from natural sources were used as active ingredients. Different types of teas, barks, herbs, spices, fruits and vegetables were selected as sources of natural antioxidants. Total phenolic and flavonoid content and total antioxidant activity of extracts were determined using spectrophotometrical methods. Obtained natural extracts were subsequently used for encapsulation. Prepared application forms were characterized in terms of their physicochemical properties. Particle size was monitored by dynamic light scattering. Colloidal stability of particles in suspension was determined using zeta potential. Spectrophotometry was used to evaluate the efficiency of encapsulation of active compounds into particles. The morphology of the new type of combined PHB liposomes was monitored by electron microscopy. Chromatography was used for quantification of individual components of particles. Morphology of nanofibers and incorporation of active agent into their structure were monitored using FTIR-ATR spectroscopy and electron microscopy. Afterwards, antimicrobial, cytotoxic and genotoxic effects of preparations were evaluated. It was found that the most suitable types of extracts for liposome preparation are aqueous and lipid extracts of natural antioxidants. Prepared particles showed excellent stability and good encapsulation efficiency. The study confirmed that incorporation of polydroxybutyrate into liposome structure does not reduce neither the colloidal stability of the particle, nor the efficiency of encapsulation process. Antimicrobial and antimycotic effect of preparations against model microorganisms Micrococcus lutues, Serratia marcescens and Candida glabrata was detected. It was found that process of encapsulation increases the inhibitory effect of natural extracts of antioxidants. The safety of preparations was assessed using two human cell cultures: epidermal keratinocytes and HaCaT cell line. Assays of cell viability and plasma membrane integrity were used to determine cytotoxicity of preparations. Low toxicity of liposome particles was confirmed by a series of cytotoxic tests. Obtained data showed that association of phospholipid with PHB polymer does not cause a significant increase in cytotoxicity in human skin cells. Genotoxicity testing on model procaryotic organism confirmed zero genotoxic potential of preparations. The new type of combined particles and polymeric fibers cant thus be used as a carrier for active ingredients, complex natural extracts, antimicrobial agents and many others.
Interaction of PHA-based particles and fibres with human cells

This bachelor thesis deals with the preparation and characterization of PHA based nanoparticles and nanofibers and their subsequent influence on the living human skin cell. Theoretical part is aimed at polyhydroxyalkanoates, preparation of PHA based nanoparticles and nanofibers and methods of their physical, biological and chemical characterization. Further, the theory of liposomes, their clasification, preparation and physical and chemical properties are discussed too. Next, the theoretical part focuses on the cultivation of human cells and general information on selected cell lines. The experimental part is focused on the optimalization of the preparation of liposomes and fibers enriched by PHA. Further, characterization of stability and interactions of these nanomaterials with human HaCaT cells was proved. Cytotoxic and genotoxic tests were performed and determination of concentration of phospholipids in prepared particles was realized too.
Preparation and testing of application forms for natural antimicrobial substances

The goal of this bachelor thesis was the study of antimicrobial and antioxidant effects of some herbal extracts: cloves, nettle, sage, chamomile, rosemary, elderberry, lavender and also lysozyme. Further, herbal extracts effect on Gram positive and Gram negative bacteria were compared. In the theoretical part were briefly characterized lysozyme, polyphenols, flavonoids and selected strains of microorganisms. In the experimental section characterization of herbs with antimicrobial and antioxidant effect was proved. Particles containing herbal extracts were prepared and encapsulation efficiency was determined. In some of tested herbs encapsulation efficiency of up to 80% was reached. Stability of the prepared liposomes was tested in the model digestive conditions and after 1 week and 1 month incubation period in model foods. Antimicrobial activity was tested on selected representatives of Gram negative and Gram positive bacteria. Gram positive bacteria were more sensitive to herbal extracts and liposomes containing encapsulated herbs, while lysozyme exhibited higher effect to Gram negative bacteria. Prepared particles with encapsulated antimicrobial extracts are especially suitable for use in the food industry.
Use of antimicrobial products of animal origin in cosmetics

The Bachelor thesis deals with characterization a whey in free and encapsulated form and its application to cosmetic industry. The theoretical part is focused on characterization of whey, especially on its antimicrobial properties, and use of this effect in selected cosmetics preparatives. Further, methods of encapsulation and particles characterization were described. In the experimental part, antimicrobial properties of whey were tested by two different methods. Whey was encapsulated into three different types of particles: liposomes, alginate and chitosan. Long-term colloid stability of particles during one and three weeks of storage in model conditions was tested. Further, particles were added into model skin creams. Long-term sedimentation stability of these emulsions was analyzed by analytical centrifuge.
Preparation of encapsulated enzymes for cosmetics application

Presented diploma thesis is focused on testing of an appropriate form of encapsulated enzymes intended for application in cosmetic and pharmaceutical industry. For encapsulation, proteolytic enzymes bromelain, papain and collagenase were used. These enzymes were encapsulated into alginate and chitosan microparticles prepared by an encapsulator and packed into liposomes. Encapsulation effectiveness was evaluated by analysis of total proteins. Particles stability was evaluated in model and real conditions by photometrical analysis of released proteins. Proteolytic activity of released enzymes in model and real conditions were observed too. Alginate and chitosan microparticles prepared by the encapsulator were found as an appropriate form of encapsulated enzymes designed to wound healing. Encapsulation effectiveness of these particles and stability in model conditions were good in comparison with liposomes. Hydrogel and water-oil emulsion were used for analysis of particles stability at real conditions. Hydrogel was found as a good option for preservation of particles as well as proteolytic enzyme activity. Emulsion made particles less stable and proteolytic activity of enzymes decreased rapidly. Encapsulation enables long-term stabilization of biologically active compounds as well as possibility of targeted transport and controlled releasing. Presented diploma thesis suggests possibilities of application encapsulated enzymes in designing more effective formulations for wound healing.
Active components of technical cannabis and their application in cosmetics

Hemp is unique plant with many therapeutic effects thanks to its large content of substances. This bachelor thesis brings information about characterization of main biological active substances in cannabis sativa, which are described in theoretical part of this thesis and determined in experimental part of this thesis. As a main active substance were determined antioxidants and cannabinoids for which the liquid chromatography method was optimized. This bachelor thesis also describes the antimicrobial effect of active cannabis substances for which it is used in the cosmetic industry. This antimicrobial activity has been studied in terms of the effect against dandruff, which is primarily caused by the high incidence of Malassezia yeasts on the skin of human hair. For the experimental part, the extracts of the technical hemp and more specific varieties of Bialobrzeskie and Finola were tested, which were further used for the preparation of the anti-dandruff shampoo. By analyzing the active substances in hemp extracts, the uniform result was obtained - the highest content of antioxidants and cannabinoids was reached by preparing of extract from dried hemp flowers. In the last part of the thesis, the antimicrobial effect of hemp extracts and prepared hemp shampoos was determined on selected strains of microorganisms and it was observed antimicrobial activity in extracts and shampoos againts Malassezia furfur yeast.
Probiotics and prebiotics and their co-encapsulation

This bachelor thesis is focused on study of probiotic cultures and options of coencapsulation with specific substrates called prebiotics. Lactobacillus acidophilus and Bifidobacterium breve were used as encapsulated microorganisms. As the specific substrates inulin, psyllium, apple fiber and hemp fiber were tested. Prebiotics was added to probiotics both in untreated form and in hydrolyzed form. The stability and viability of the cells have been studied by microscopy for the period of 6 weeks. Particles were stored in the enviroment of 2.5% citric acid. As the best substrate for cell growth the hemp fiber was found. The hemp fiber exhibited the highest increase of cells count. During observation of long-term stability in the model digestive conditions, particles with hemp fiber were destroyed in instestinal juices. For that reason, particles with hemp fiber could be used in food supplements with targeted transport. The bachalor thesis is also focused on characterisation of prebiotics components. The highest amount of total sugars in the group of non-hydrolyzed prebiotics exhibited psyllium. The highest content of reducing sugars both in the groups of non-hydrolyzed and hydrolyzed prebiotic had hemp fiber. By the method of thin-layer chromatography was found out that the main components of hydrolyzed prebiotics are glucose and fructose. Further, some disaccharides and oligosaccharides were obtained by TLC analysis. For characterisation of prebiotics by HPLC the column Rezex exhibited better sensitivity. HPLC analysis confirmed that the main components of prebiotics are glucose and fructose, some more complex carbohydrates were found too.
Characterization of cosmetic dispersion by centrifugal analyser

Cosmetic products are widely used nowadays. These products are mostly emulsion systems, in which processes that lower physical stability can take place. This can leads among others to the shortening of the expiration date or to the reduction of the efficacy of the product. So, in the cosmetic practice, the physical stability is monitoring by tests. There are more types of these tests and they can be realized under different conditions. This Bachelor Thesis is focused on monitoring the physical stability of samples of cosmetics products by using an analytical centrifuge LUMiSizer and by using methods that are commonly used in a cosmetic practice and development. The aim of this study is to compare single results and to verify the suitability of the use of LUMiSizer for accelerated determine of the physical stability of cosmetic products. The stability of samples over time and the effect of the temperature on the stability of samples were monitoring by using the LUMiSizer device. Freeze-Thaw cycles and the conventional method of testing the stability of samples were applied as well. There was also determined the effectiveness of the stability test, mentioned by the literature, in this work. Based upon the results of experiments that were carried out and upon information given by manufacturers of samples that were used, the LUMiSizer device was evaluated as a useful tool for assessing the stability of samples although results obtained by the LUMiSizer device are not exactly the same as results of the conventional method. Although the application of additional methods is recommended.
Viability of probiotic bacteria in some foods

This bachelor thesis is focused on viability od probiotic bacteria in some foods and looking for optimal food and beverages, that exhibited a minimal negative effect on the growth and viability of probiotic bacteria. Lactobacillus acidophilus, Bifidobacterium breve and commercially complex of probiotics were tested. In the practical part, probiotics were incubated in the environment of selected types of model and real foods. Further, incubation in model gastrointestinal juices to simulate human digestion was performed. Model foods with a moderate value of concentrations of sugar, protein and alcohol were shown the best alternative for growth of bacteria and their lowest mortality. For salt, the smallest concentration was shown as the most suitable. The chicken broth, chicken and beef meat were evaluated as the best environment for the capsule with complex probiotics. The vegetables and vegetable broth were stated like the best for Lactobacillus acidophilus and Bifidobacterium breve. According to the preliminary results we can conclude that the probiotics should be preferably consumed during meals and with the sufficient amount of liquids for the activation of all cells.
