Norway spruce chloroplast ultrastructure - heterogeneity within a needle.

6 Abstract Temperate forests serve as long term carbon storage and are affected by increasing carbon dioxide (CO2) concentration in the atmosphere. Norway spruce (Picea abies (L.) Karst.) is the most abundant conifer in the forests of the Czech Republic, therefore I studied the response of its photosynthetic aparatus to elevated CO2 concentration. The aim of my thesis was to analyze the impact of CO2 concentration on chloroplast ultrastructure in both shaded and exposed needles, focusing on the volume density of starch in the median cross-sections of mesophyll cell chloroplasts. The next aim of the study was to test whether the chloroplasts of the first subepidermal layer of mesophyll are representative for the whole needle with respect to starch volume density. The study was performed on eleven years-old Norway spruce trees that had been exposed to ambient or elevated concentration of CO2 for six years; the experiment had been carried out at the Bílý Kříž experimental station in the Beskids Mountains in cultivation chambers with automatically adjustable windows. Norway spruce (Picea abies L. Karst.) is an abundant conifer tree in European temperate and boreal forests, which behave as carbon sink in the global carbon cycle. The physiological response to elevated CO2 concentration may be interconnected with changes in leaf anatomy and morphology. Changes in anatomy of coniferous needles in response to anthropogenic pollution

The anatomical structure of conifer needles is affected by external enviromental conditions. From abiotic factors, apart of irradiation, main determining factors are drought and extreme temperatures belonging to factors of climate change. Anthropogenic pollution has a significant position among the external abiotic factors that cause changes in the structure of needles. At present, the main polluting compounds are tropospheric ozone, SO2, NOx and PM. Due to the effect of atmospheric pollutants, the proportion of individual tissues in the needle changes, so does the proportion of intercellular spaces and the internal structure of cellular organelles, especially chloroplasts. Mesophyll cells show signs of degradation, especially around the substomatal cavity. There appears a visible damage on the needle surface - chloroses and necroses. The process of senescence speeds up during the activity of stress enviromental factors, such as anthropogenic pollution. The main objective of the present Thesis is to gather knowledge about changes of conifer needle structure, caused by anthropogenic pollution, especially ozone, SO2 and NOx, in combination with climate change factors - drought and extreme temperatures. Vliv solární radiance na strukturu xylému jehlic smrku ztepilého

In this thesis is evaluated the xylem structure of Norway spruce needles depending on solar radiation and altitude. Same-age spruce stand has been selected on the area of Hruby Jesenik at different altitudes on the southern and northern exposure. Sun needle samples were taken on each stand. The analysis of xylem structure was done. The number of tracheids, tracheid diameter, tracheid area and xylem area were evaluated. It was found that more than 50% dependence of xylem parameters on altitude and solar radiation, where exposure significantly affects the xylem structure only at altitudes above 1 200 m. The prediction maps show that the highest values of predicted xylem parameter values are found between 800 m and 1200 m about sea level.

Metallurgy along the East Aegean-West Anatolian Interface in the Second Millennium B.C.

(in English): This thesis aims at collecting, cataloguing and analysing bronze objects from the area of the East Aegean-West Anatolian Interface in the second millennium B.C. Based on closer typological assessment and comparanda, the question of eventual local specific production along the Interface, different from the Aegea or Eastern Mediterranean, is being investigated here. From up to 217 collected items, indeed many types of bronzes, especially swords, razors and spearheads indeed show a set of specific features. Also, the characteristic of bronze metals differs in Lower Interface with stronger minoan-mycenaen influnce from items in Upper Interface which seems to be following more anatolian features.
Bone pins of the Early Bronze Age in Bohemia and Moravia

Bone pins of the Early Bronze Age are interesting, not too frequent, and still properly unprocessed kind of artifact. Probably based on an older tradition, but they were soon replaced by the metal pins. The main objective is to develop as most detailed catalog of findings as is possible, tracking the finding context and chronological position of bone pins in the Early Bronze Age in Bohemia. The subject of the work is also a question of technology used in the production bone and antler industry and its evidence from the Bronze Age, focusing on the technology and processes used in the production of bone pins. Another factor which is monitored is the social status of bearers of the bone pins in the Early Bronze Age.

The effect of oribatid mites on the composition of fungal community in pine litter

4 Abstract The aim of this study was to determine whether oribatid mites affect the variety of fungi in litter of Scots pine (Pinus sylvestris), and if the dispersibility of fungi particles is related to oribatid mite species. To this end, I have prepared several semi-natural systems, in which I observed dispersion of fungi separately and in the presence of two oribatid mite species: Phthiracarus sp. and Adoristes ovatus. I assessed the experiment in two ways, by a cultivation of washed needles on agar medium and direct observation of needles under a binocular magnifier. In the next part of this study I isolated particles of fungi present on the surface and in the faeces of oribatid mites Adoristes ovatus, Porobelba spinosa and Spatiodamaeus verticillipes and compared it with the respective oribatid mite species. The most frequent needles fungi spread by oribatid mites were Oidiodendron maius var. maius, Oidiodendron muniellense, Oidiodendron setiferum, Umbelopsis ramanniana, Mucor hiemalis f. hiemalis and Mortierella hyalina. On the mites body surface the most commonly recorded fungi were Beauveria bassiana, Umbelopsis ramanniana, Cladosporium cladosporioides s.l., Cladosporium herbarum s.l. and unknown species of the genus Penicillium.
