Subunit c of mammalian F1Fo ATP synthase - from molecular mechanisms of assembly to potential therapies

Mammalian F1Fo ATP synthase plays a crucial role in ATP production through the process of oxidative phosphorylation. Assembly of this multisubunit protein complex requires specific assembly factors. Notably, the assembling of subunit c in mammals requires several factors, yet despite decades of research, the import of subunit c, its incorporation into the inner mitochondrial membrane, and the assembly of monomers towards octameric c-ring remain enigmatic. To shed new light on this process, we first screened for interacting partners of subunit c using a mass spectrometry-based approach. Our screen identified three proteins as the most prominent interactors of subunit c- TMEM70, TMEM242, and c15orf61, which we subsequently characterized. For the initial characterization of the TMEM242 function, we generated both knockdown and knockout HEK293 models. TMEM242 knockdown led to impaired biogenesis and decreased levels of assembled ATP synthase without affecting the content of other OXPHOS complexes. On the contrary, complete deficiency of TMEM242 lead also to the downregulation of complexes I and IV, which indicates that the primary target of TMEM242 is ATP synthase. The content of my bachelor's thesis is the solution for the implementation of a prefabricated reinforced concrete skeleton and its cladding. The work includes a technical report, site plan with broader transportation connections, bill of quantities, technological specification focusing on skeleton assembly, construction organization solution, schedule and workforce graph, machinery layout proposal, inspection and testing plan for the prefabricated skeleton assembly phase, safety and health plan for the construction site, budget for the specific phase, details of connections for the prefabricated structure and enclosure, and transport scheme.
Production Process Improvement

This bachelor thesis focuses on the analysis of the production process at HydrauliCS s.r.o., with the aim of streamlining the hydraulic cylinder production process using selected lean methods. The first part describes the theoretical background to introduce the reader to the issues of this thesis. In the second part, the product representative, the current state of its manufacturing process, and a detailed analysis of the assembly process is presented. For the analysis, the work analysis and measurement method, and spaghetti diagram are used. In the last part, suggestions for eliminating the deficiencies in the specific process and their economic evaluation are presented.
Optimization of a machine assembly process

The diploma thesis deals with the optimization of the assembly of a machine designed for printing paper material. In the introduction, the company Windmöller & Hölscher, with which the work was collaborated, is introduced. The theoretical part is focused on lean production methods and types of industrial printing. In the practical part, the design is optimized with the use of lean production, adjustment of the assembly procedure, creation of instructions and adjustment of the layout of the assembly box. The proposed optimizations are subsequently evaluated from a financial point of view and, if possible, the return on investment is calculated.
Woodworking machines for furniture industry

The bachelor thesis deals with woodworking machines that are used in the furniture industry to produce furniture parts. Initially, this topic describes wood as such. It discusses in detail its structure and composition, which are important in determining the physical and mechanical attributes of wood. The topic then focuses on the main machining methods by which wood can be processed, including the machines used for this purpose. Problems in sawing wood, that are caused by defects in the tool or in the material being machined, are mentioned. There is also a brief classification of furniture products according to the furniture standard, where furniture is classified using several factors. Finally, all the woodworking machines and methods are interpreted on the design of a wine rack, the manufacture of which is discussed in detail from the initial collection of the semi-finished product to its final assembly. A technological process is drawn up for all the parts needed to assemble the stand, accompanied by drawings.
Additive manufacturing of fixtures for assembly and pressing by FDM

This bachelor thesis dealt with the design of fixtures for prototype assembly of thermoregulating modules. One of the aims of the thesis was to evaluate the suitability of using FDM printing for the production of prototype fixtures. FDM fixtures are intended to replace the metal fixtures used to date. The motivation for the use of additive manufacturing was to make production cheaper and faster within the prototype department of Schaeffler Production CZ, s.r.o. The effect of the surface loading on the 3D printed PETG material, which was chosen as the construction material within the scope of the work, was measured. The results of this measurement and the findings from the research part were used for FEA analysis and design of the product. The comparison of the cost-effectiveness and the speed of the prototype fixture production clearly shows that the use of additive technologies is better.

In order for the product to remain on the market, it tries to combine the principles of solitaires with modular systems. Cardboard is intended as the final material. The work addresses the design, ergonomics and aesthetic appearance in the context of purchasing potential.
Analysis of single-cell genomic data of Saccinobaculus sp.

The progress of single-cell genomics and metagenomics techniques allows us to explore the uncultured organisms in greater depth. In this work, we focused on the genome assembly and genetic code of the oxymonad Sacinobaculus ambloaxostylus, which inhabits the hindgut of wood-eating cockroaches from the genus Cryptocercus. Using single cell picking and whole genome amplification we obtained sequencing data from three cells of S. ambloaxostylus. The obtained data was used to generate a single-cell genome assembly using SPAdes. The assembly was further binned and decontaminated to remove any potential bacterial or eukaryotic contaminants. After decontamination and re-assembly, we obtained a genome assembly with a total length of ∼417Mbp distributed to ∼185 thousand scaffolds. Despite its low completeness of 13.71%, we attempted to determine the genetic code used by S. ambloaxostylus. For this we manually modeled 33 genes from various metabolic pathways. Aligning these gene models with homologs from closely related oxymonads, we calculated the usage of stop codons as sense codons with focus on conserved positions. Woodworking machines used in furniture industry

This bachelor thesis deals with the characteristic of woodworking machinery, which is used in creation of furniture. The first part contains the brief description of the wood and wood-based materials and their properties. The second part deals with the own decryption of wood-working machinery and its use in furniture manufacturing. The last part specifies the furniture industry and the basic division of furniture. As part of this thesis, author`s design of coffee table was developed as the showcase of the furniture-manufacturing and presentation of the machinery, tools including the assembling.

The annexes contain drawings of the construction site facilities and the situation of the construction with broader transport relations.
