Stability of Soil Organic Matter

This bachelor thesis summarizes available information about current research of soil organic matter stability and it divides them into two logical units. The first part includes environmental, physical and chemical factors, including human activities, which affect the stability or decomposition of soil organic matter. The methods used for the study of organic matter in the soil are summarized in the second part of this thesis. There the attention is paid to the accumulation of organic matter in specific pools, the ways of extraction of organic compounds and the use of quantitative and qualitative analytical methods to describe various characteristic of organic matter.

Importance and impact of phthalates during peritoneal dialysis and current possibilities of their replacement

This thesis deals with the importance and influence of phthalates in peritoneal dialysis (PD) and the current possibilities of their replacement. The information about phthalates – esters of phthalic acid - was completed with focus on the most commonly used plasticizer in medical material, bis(2-ethylhexyl) phthalate (DEHP) and its effects on the human organism and the environment. The possibilities of substitution of PVC in peritoneal dialysis were searched and their advantages and disadvantages were evaluated. As the evidence of phthalates absorbed by the human body in carrying out peritoneal dialysis selected studies performed in Germany and in our country, dealing with this issue, were used. High performance liquid chromatography (HPLC) with UV detection at 224 nm or gas chromatography - mass spectrometry (GC / MS) was used here for the determination of phthalates in the blood and dialysate in peritoneal dialysis. Extraction of sample with the suitable solvent and purification of the extract predates the actual analytical determination of sample.
Methods for study of plant phosphoproteins

Reversibly phosphorylated proteins play a role in many fundamental cellular processes. An example is the cytokinin signal transduction pathway in plants that is mediated by the so-called two-component system involving histidine and aspartate phosphorylation. Due to the very low stability of phosphorylation of these amino acids, is difficult to find a suitable method for phosphorylation studies. The theoretical part of this bachelor thesis describes not only the current experimental approaches for analysis of protein phosphorylation in two-component systems but also is trying to propose new approaches for this analysis. There are described methods based on labeling of proteins using [?-32P] ATP and on the detection of released phosphate employing the colorimetric method or ICP-MS. Another possibility is sensitive identification of phoshorylated and non-phosphorylated forms of protein fused with GFP using CE-LIF technique. The practical part of the thesis deals with optimization of the selected colorimetric methods and also is focused on the preparation of the AHP5 protein fused with GFP for CE-LIF analysis.

Analysis of low abundant lipids in vernix caseosa using chromatographic methods and mass spectrometry

The aim of this work is to develop a separation method for characterization of unknown lipids in vernix caseosa. We focused on very low abundant lipids which have not been previously described in vernix caseosa. The total lipid extract was fractionated using low pressure normal phase liquid chromatography to obtain sub-fractions. The sub-fractions were further separated in high performance liquid chromatography mode to isolate classes of lipids. This procedure enabled us to characterise lipid classes by mass spectrometry and identify one of the class as acyl-ceramides with non-hydroxylated fatty acids.
Establishment of method for determination of selected triazine pesticides in water using GC-MS/MS

This bachelor thesis deals with the determination of triazine pesticides in water using GC-MS/MS. Firstly, the development and optimisation of an analytical method was performed. The following parameters of the analytical method were chosen for optimisation: carrier gas flow rate, temperature gradient, selection of SRM transitions and collision energy of SRM transitions. Then, several extraction parametres such as SPE column drying time, selection of elution solvent, sample pH and sample volume were optimised. Finally, the method was successfully validated and applied to the analysis of a real tap water sample.
Separation of nonpolar lipids from vernix caseosa

The aim of this work is to develop a separation method for characterization of unknown nonpolar lipids in vernix caseosa. Here we optimized adsorption chromatography coupled to mass spectrometry detection with exact mass measurement. A long silica gel column (250 + 250 x 4.6 mm, particle size: 5 mu m) provided the best separation of nonpolar lipids present in vernix caseosa.
