Cardioprotective effect of adaptations to hypoxia and cold: Connexin 43 and hexokinase 2

Ventricular arrhythmias are the main cause of death worldwide. An increased incidence of arrhythmias in the heart of mammals is accompanied by a remodeling of the cellular distribution gap between the channels of mainly connexin 43 (Cx43). Recently has been demonstrated significant effect of mitochondria and their association with arrhythmogenesis. Various pathological conditions alter the expression and / or distribution of Cx43, depending on the phosphorylation status. but also on altering the association of hexokinase with mitochondria, which reduces the likelihood of apoptosis activation. Adaptation to intermittent hypobaric hypoxia potentiates endogenous pathways reducing the incidence of ventricular arteries, whereas continuous normobaric hypoxia does not have this effect. Another studied model is cold acclimatization, which has been known for several decades by known effects on human health. However, the heart study of these models in relation to Cx43 is missing. Our goal was to determine the expression of Cx43, phosphorylated Cx43 (p-Cx43) and hexokinase (HK1, HK2) and their distribution in cardiomyocytes. The effect of acute cold and permanent light to left ventricular of the rat heart

Acute cold exposure increases the risk of sudden cardiac events, similarly exposure to constant light negatively affects the cardiovascular system. However, the individual effects of these factors and the effect of their combination on cardiomyocytes are not yet known. The thesis deals with the influence of a 3 day cold exposure and constant light on the expression of β-adrenergic receptors and associated G-proteins in association with apoptotic signals in the left ventricle of the Wistar rat heart. In this work apoptotic proteins BAX, BCL2, caspase 8 and important components of β-adrenergic signalization - β1, β2, G-proteins, Gas, Gi1/2 and Gi3 were determined. The relative expression of the proteins was analyzed by Western blotting. The results confirm the detrimental effect of cold and light exposure. However, the synergistic effect of these two stressors shows surprising results.
Energy supply for greenhouse production

The import of the vegetable has prevailed in the long run over the own production. In connection with this reality and increasing demand for the food with higher quality the production greenhouse is more often built. The providing heat, cold and electricity for the year-round operation is related to the construction and working of these objects. The theoretical part of this theses describes possible ways how to cover energy consumption for the model greenhouse in Kožichovice, Třebíč region. The heat losses and the year-round heat consumption were stated for the selected model and compared with the exact values from Kožichovice. The following method of energy supply was selected based on the calculated energy consumption - the separate cogeneration unit, cooperation of several cogeneration units, a biogas station, a heat pump and central heating system. These methods of energy supply were subjected to technical and economical comparison.

The effect of season on mitochondrial swelling of rat heart after cold acclimation

Risk of death from cardiovascular disease is substantially increased throughout winter season and by exposure of organism to acute cold stress. However, several studies have suggested that some cold acclimation models could act cardioprotectively. The thesis is focused on effects of acute and chronic cold acclimation on swelling of rat heart mitochondria. Mitochondrial swelling is, besides other things, caused by massive influx of Ca2+ into the mitochondria especially during ischemia-reperfusion injury. By that, mitochondria permeability transition pore is being opened and subsequently, this could end up in mitochondrial burst and cell death, eventually. Lower rate of mitochondrial swelling indicates greater mitochondrial resistance. The results express higher mitochondrial resistance after acute cold exposure and chronic cold acclimation. The bachelor thesis was focused on the physiological, cellular and transcriptomic characterization of Arabidopsis thaliana treated with cold under different light conditions. To determine the role of the cold stress in leaf ontogenesis, the analysis included leaves in the different developmental stages. It was found that 4 weeks old plants have almost completely developed 6th leaf and cold treatment has no impact on its cellular morphology. In contrast, younger leaf number 14 which initiates a transition from proliferation to expansion stage was strongly affected by cold treatment. It is shown that exposition to a low temperature significantly reduces its cell area and a number of epidermal cells. Propidium staining confirmed that young proliferating leaves are also more sensitive to stress conditions than older leaves. The higher sensitivity of young leaves to cold stress was accompanied with lower expression of CBF genes that play an important role in cold stress tolerance. Younger leaves also showed higher expression of cytokinin responsive genes and genes related to cell cycle corresponding to ongoing developmental processes.
Relationship between the renin-angiotensin-aldosterone system and the heart and its influence during 5°C cold acclimation.

Heart as one of the life-important organs is influenced by many ongoing processes in organism. Even the smallest molecular changes can importantly influence heart's physiology and activity. Last years, there were many studies concentrating on clarification of renin-angiotensin- aldosteron system (RAAS) and its influence on heart and kidneys. Cardiovascular diseases are usually caused by changes in RAAS and that is the reason why more and more experts are examining the extent to which diseases can be prevented and heart function improved. Ambient temperature is one of the factors that greatly affects the RAAS and the heart. In this thesis I tried to summarize current knowledge about RAAS and its effects on organism, focusing on influencing the physiology of the heart. While ambient temperature has significant effect on the RAAS and the heart, this thesis points to the knowledge associated with cold acclimation at 5 řC. Algal and Cyanobacterial Adaptations to Low Temperature and Desiccation

Algae and cyanobacteria, due of their evolutionary antiquity, are widely distributed primary producers that can withstand extreme environmental stresses. Low temperature, freezing and melting, and desiccation and rewetting, are common stresses prevalent mainly in polar regions and in winter seasons of temperate areas. In terms of physiological effects, these types of stresses share similar effects or are closely related to one another. Low temperatures and desiccation exert a variety of stresses that need to be negated or lessened by adaptations. Specifically, adaptations to chill, freeze, and desiccation stresses will be discussed, as well as strategies that allow for stress avoidance or resistant morphological adaptations. In this thesis, characteristics, functions and mechanisms of these adaptations and stresses are reviewed, as well as potential biotechnological uses of said adaptations.
