Composting of urban green areas in urban compost facility in Březnice

This thesis deals with the use of municipal biowaste, which processes compost facility in the city Březnice. In this work are dealt optimal conditions for composting, composting grass biomass then composting technology and at the end concept of composting in the city Březnice with a total processing technology and equipment. The aim of my thesis was to outline the composting and propose optimal solutions for the construction of a composting facility. Part of this work is also an experiment with ion exchange capacity, which proves the importance of following the correct composting process for the production of quality compost. The contribution of this work rests primarily as such its integrity and organization. It is possible to find there the right parameters for creating the right conditions of the composting process, including the elements necessary for actual registration of compost.
ENDOCRINE DISUPTIVE CHEMICALS: MECHANISMS OF WHITE ROT FUNGI BIODEGRADATION

The main aim of this study was to elucidate mechanisms involved in EE2 and other EDs compounds degradation by white rot fungi. Results showed simultaneous involvement of ligninolytic enzymes and cytochrome P-450 in biodegradation. The present results indicate the advantage of utilization of WRF cultures for organopollutants degradation.
Functional thin films for applications using advanced oxidation processes

This diploma thesis aims to optimalization the process of magnetron sputtering and creating of thin layers for use in advanced oxidation processes. During the work was created range of TiOx layers. For this process was used physical method of sputtering called PVD. The photocatalytic activity of the deposited films was tested by degradation of organic dyes Acid Orange 7. Furthermore, the layer was analyzed on surface morphology (SEM) and the layer thickness (profilometry). Study of created layers was focused on the link between the characteristics of each layer, deposition parameters and photocatalysis properties. Based on these results, the layers were applied in a system using AOP for the decomposition of organic substances. The first chapter is devoted to a summary of existing knowledge of photocatalysis and its principles. Another chapter is devoted to the theory and methods of applying thin layers and summary of knowledge of the low-pressure discharges. In the exprimental section there are described various components of the apparatus. Furthermore, the experimental part of the work focuses on the analysis of the optimization process of applying thin layers on titanium oxide. The last chapter of the thesis contains the results of the experiments on the basis of is designed another research progress of this issue.
The Present and the Desirable Future of the Waste Treatment and Utilization in .A.S.A. Lišov

The subject of these theses contains an evaluation of the waste policy in the A.S.A. České Budějovice company. More precisely is oriented on waste dump in Lišov. Firstly, as an result of my work, the proposition of better economic and technology solution is offered. The proposal covered also the problem of the ecological waste handling. In the frame of the ecological treatment the biodegradation is included. This will lead to another direction of the waste treatment policy. The way is under EU environmental policy legislature. The final result of these theses is a real project of the waste assorting line with the direct link towards the manipulation with the biodegradation material. These procedures are proposed in accordance with the economical situation of the A.S.A. České Budějovice division.

The efficiency of bioremediation depends on a number of factors, the most important of which are the ability of the bioremediative organism to degrade the target pollutants, the pollutant’s intrinsic properties (aqueous solubility being of key importance), and soil parameters. Comparison of in vitro tests for measurement of estrogenity of chlorinated compounds and their use in the evaluation of biodegradation of chlorbenzoates by the fungus Panus tigrinus

Recently, contamination of the environment by endocrine disruptive compounds, toxicity of these chemicals and their biodegradability are widely studied. This work is focused on estrogenity of chlorinated phenols and chlorbenzoic acids that represent potential degradation products of PCB compounds. Two different yeast bioassays for evaluation of estrogenity based on recombinated strains of Saccharomyces cerevisie were compared in this work. Estrogenic activity of chlorbenzoic acids varied with the chemical structure of chemicals. A mixture of chlorbenzoates (initial concentration 10 ppm) was degraded by a ligninolytic fungus Panus tigrinus in liquid cultures. Chemicals resulting from degradation were tested for their estrogenity, however, results indicated that the test responses were inhibited probably by some compounds present in the fungal culture liquid

The aim of this study was to evaluate the mutagenicity of three mixtures of synthetic dyes used in textile industry. Detection of mutagenicity was carried out with three samples containing residues of textile dyes obtained from textile company Jitex Písek. Two different biodegradation process were used for elimination of genotoxicity, biodegradation using the bacterial activated sludge and the enzyme systém of the ligninolytic fungus Irpex lacteus. The genotoxicity of the samples was detected by the Ames test with and without metabolic activation in vitro, using S9 liver microsomal fraction and cofactor mixture. Salmonella typhimurium His- strain TA98 was employed for detection of frameshift mutations in the Ames test. During two different biodegradation processes (using bacterial activated sludge and the ligninolytic fungus Irpex lacteus), the change of mutagenicity of the degradation products of dyes was monitored
