Molnár, Jakub ; Čáslavová, Eva (advisor) ; Tejkal, Karel (referee)
Title: Management and marketing of the project Sports talent of the year and its implementation for period 2014-2015 Targets: The aim of this thesis is to propose improvements of the project Sports talent of the year for period 2014 - 2015. The project is focused on healthy lifestyle of the primary school pupils in the Czech Republic. The author based on analysis of previous years which were realized by a profit organization. His own contributions are made by the implementation of the project. Methods: There is a case study used to analyze the information about the project Sports talent of the year. Analysis of the conditions is carried out with the aid of PEST analysis, Porter's model of five forces, and analysis of resources. SWOT analysis is applied to synthesize previous analyzes. Results: The study revealed gaps especially in the management activities of the project Sports talent of the year. The greatest gaps can be seen in the branch of staffing, marketing and communications and financing. Key words: sports event, project, non profit organization, SWOT analysis, management and marketing of sports event

Circadian regulation of glucocorticoid secretion
Tejkal, Karel ; Sumová, Alena (advisor) ; Soták, Matúš (referee)
Glucocorticoids belong to a family of steroid hormones synthesized in the adrenal gland. They fulfill a variety of functions related to metabolism, immune response and ontogenesis. Glucocorticoids function as the end-effector of hypothalamic-pituitary-adrenal (HPA) axis and as such, their levels in blood are elevated after exposure to stressors. The basal levels of glucocorticoid also show a pronounced diurnal rhythm, suggesting involvement of the circadian clock in the regulation of HPA axis. Studies have shown that other regulatory mechanisms apart from the HPA axis are involved in regulation of diurnal glucocortioid secretion. The disturbances of the regulatory mechanisms may lead to serious pathological conditions. This thesis describes the rhythmic nature of glucocorticoid release and mechanisms by which the circadian clock exerts its influence over the rhythm. Thereafter, the feedback of glucocorticoids onto the clock system is briefly explained. Finally, some examples of a role of abnormal glucocorticoid secretion in selected pathologies are provided.
The role of glucocorticoids in circadian system
Tejkal, Karel ; Sumová, Alena (advisor) ; Forman, Martin (referee)
Glucocorticoids are mammalian steroid hormones secreted from the adrenal gland. The basal levels of glucocorticoids show a pronounced diurnal rhythm with maximum at the beginning of the active period and minimum at its end. Glucocorticoids have an influence over a variety of metabolic functions and their secretion is tightly regulated. This regulation also depends on the circadian system, which utilizes glucocorticoids to entrain the peripheral tissues by inducing rhythmic gene expression. The mechanisms by which glucocorticoids influence mammalian circadian system has not yet been precisely defined, especially concerning the influence of glucocorticoid signalling on gene expression in different tissues and the dynamics of glucocorticoid receptor (GR) occupancy. This thesis studies the influence of ablation of glucocorticoid signalization induced by adrenalectomy on the clock gene expression of in the central clock in the suprachiasmatic nucleus and peripheral clocks in the hippocampus and distal colon. The effect of adrenalectomy on gene expression is compared with the effect of restricting the feeding time, which has also been shown to affect glucocorticoid levels in the body. Other experiments were aimed at elucidating impact of changing the activity of GR on gene expression using synthetic GR...
