Utilization of polymerase chain reaction technique for identification of probiotics in Makgeolli rice drink

The Bachelor’s thesis is divided into two parts, namely theoretical and experimental. The theoretical part provides a definition of probiotics, representatives of these probiotic microorganisms, and their use in clinical practice as well as in the fermentation process. Furthermore, the mechanisms of action of probiotics in the organism are described. In the experimental part, the presence of probiotics in commercial and homemade makgeolli products is analyzed using polymerase chain reaction (PCR).
Evaluation of the presence of probiotic bacteria in a food supplement using molecular biological methods

Probiotics are beneficial microorganisms that, when administered properly, can provide health benefits to the host, such as aiding digestion and boosting the immune system. Their effectiveness and benefits depend on the specific strain of microorganism. These strains are often from the genera Lactobacillus, Bifidobacterium, and represent natural hosts of the gastrointestinal tract. This bachelor thesis focuses on the identification of probiotic bacteria in a commercially available dietary supplement Linex® Forte using molecular biological methods, mainly polymerase chain reaction (PCR). The aim was to verify the presence of the claimed probiotic cultures of Lactobacillus acidophilus, Bifidobacterium animalis, and to compare the results with the manufacturer's claims. Therefore, two DNA isolation methods were used: phenolchloroform extraction and the commercial purification kit OMNI International. The isolated DNA was then analyzed by domain, genus and species-specific PCR. The results show that the methods used are suitable for the identification of probiotic bacteria and confirm their presence in the selected product.
Utilization of PCR technique for identification of probiotic bacteria in daily hygiene product

Probiotic bacteria have traditionally been used in the food industry, but their applications have now expanded to the prevention and treatment of various diseases. Increasing evidence supports the efficacy of bacteria of the genus Lactobacillus in the oral cavity has led to the application of probiotic strains of this genus in mouthwashes and other personal care products. In this bachelor thesis, DNA was isolated by two methods, then quantified spectrophotometrically and amplified via conventional PCR. The results of the polymerase chain reaction were detected by gel electrophoresis. The presence of probiotic bacteria species Lactobacillus delbrueckii, Lactobacillus plantarum and Lactobacillus pentosus was confirmed.
Verification of the presence of probiotic bacteria in a cosmetic product using the polymerase chain reaction (PCR) technique

This bachelor thesis focuses on the verification of the presence of probiotic bacteria in a cosmetic product using the polymerase chain reaction (PCR) technique. The theoretical part provides an overview of probiotics, their application in cosmetics and their effect on skin condition. Furthermore, the most commonly used molecular biological methods for the identification of probiotic organisms are described. In the practical part, the work focuses on the isolation of bacterial DNA from cosmetic products and its subsequent amplification by PCR. The amplified products were analyzed by agarose gel electrophoresis and detected by UV irradiation. The results showed the presence of bacteria of the genera Lactobacillus and Bifidobacterium, but the presence of bacteria of the genus Lactococcus, which were declared by the manufacturer together with the above genera, was not confirmed.
Molecular identification of probiotic bacteria in milk products form commercial yoghurt cultures

Lactic acid bacteria are considered as an important group of bacteria with probiotic effects, which are being widely used in the food industry or pharmacology. Identification and characterization of important probiotic strains play an essential role in the validation of probiotic products for commercial purposes. Their identification using molecular-biology techniques (most commonly PCR method) is one of the standard tools in commercial operations and services. The aim of this bachelor thesis is a literature review of probiotics and probiotic strains as well as a summary of current knowledge about the use of molecular biology techniques for identification of these bacteria with probiotic properties in dairy products. The experimental part of this work verifies the presence of probiotic bacteria declared on selected commercial dairy products using the polymerase chain reaction (PCR) method.
Detection of probiotic bacteria in diary food products using PCR technique

In the bachelor thesis, DNA was isolated from commercially available white yogurt. The isolated DNA, which was gained by two different methods, was performer analyse by a spectrophotometer. Both methods provided sufficiently concentrated and high-quality DNA for further analysis by PCR. Precisely defined sections of isolated DNA were amplified using specific primers. The presence of the bacterium domain was detected, and in the case of genus specific amplification, the presence of bacteria of the genus Lactobacillus was detected too, by gel electrophoresis.

Probiotic bacteria play an important role in a healthy lifestyle. They help the consumer maintain the balance of intestinal microflora and prevent the overgrowth of harmful bacteria. Accurate identification and characterization of these probiotic strains is essential for research and the food industry. For exact identification, the use of molecular biological methods is necessary, thanks to which it is possible to validate probiotic products for commercial use. In this bachelor thesis, the DNA of probiotic bacteria was isolated from available dairy products. Two methods of isolation were used to isolate bacterial DNA, both of them provided sufficiently concentrated and high-quality DNA for further analysis by polymerase chain reaction (PCR). The presence of the Bacteria domain, genera Lactobacillus and Bifidobacterium were proved. Finally, the presence of Lactobacillus acidophilus species was also detected in the products.
Validation of probotic bacteria presence in fermeted plant food products

Probiotic bacteria are an essential part of human nourishment which provide many health benefits. We consume them not only in traditional dairy fermented foodstuff, but also in the ever-expanding fermented plant products. The theoretical part summarized general knowledge about probiotic microorganisms, individual species used in technology of processing plant and dairy fermented products, and their influence on human health. The following chapters are devoted to various methods of identification of probiotic bacteria with emphasis on molecular diagnostics, specifically by PCR method. The experimental part is focused on the identification of lactic acid bacteria in a plant fermented product – in a fermented cabbage using 2 methods of isolation bacterial DNA and subsequent analysis by PCR method.
Identification of probiotic bacteria in cosmetic creme using PCR technique

Probiotic bacteria are found in a wide range of food products and their internal use brings many positive effects for the human body. Thanks to these benefits, probiotic bacteria began to be used in the cosmetic industry with the assumption of similar positive effects as with internal administration. The aim of this bachelor's thesis was the theoretical part, which was devoted to various genera of probiotics, which are most often used in cosmetic products, in which skin diseases, surface probiotics are most often used and what effect they bring in treatment. Subsequently, the most commonly used molecular-biological methods for the identification of probiotic bacteria were described. In the experimental part, the DNA of probiotic bacteria declared by the manufacturer was isolated from two cosmetic creams and subsequently their presence was verified using the polymerase chain reaction method.
Power and Temperature Coefficient During Nuclear Power Unit Operation

This master thesis deals with the possibilities of traffic of nuclear power unit at thermal and power effect at the end of the campaign, focusing on VVER reactors. For a better idea of the reader the design of key components of the unit in terms of performance is analysed. Parameters of relevant components for Dukovany NPP are presented briefly. The possibilities of traffic of nuclear power unit on thermal and power effect at the end of the campaign are particularly demonstrated on the example of the Dukovany NPP. Furthermore the program Moby-Dick is introduced and the basic possibilities for its use to calculate the course of the campaign are described. At the end of the thesis, we conducted sample calculations for the duration of the campaign on the fourth block of the nuclear power plant.
