Automatically Updated Web Portal

This bachelor's thesis is dedicated to the design and implementation of an automatically updated web portal that tries to resolve the shortcomings of the portals filled with other people's content. Furthermore, it presents a comparison of the existing scientific portals, it discusses the problems of extraction, saving and searching for information. General mechanisms are demonstrated on the European research projects portal, which removes the shortcomings of CORDIS, the official information portal for European research and development. The thesis takes the existing product as a prototype and its aim is to improve the quality of the extraction and extend the system to detect any potential problems and notified an administrator of them. This was achieved by increasing the robustness and speed of the extractor, by registering all the important events associated with the extraction and, on the other side, the implementation of the separate administrator section of the web portal, which informs the administrator about problems and offers the problem-solving devices.
The role of ETV6-RUNX1 fusion protein in the sensitivity of leukemic cells to L-asparaginase

Translocation t(12;21) with the presence of the fusion gene ETV6-RUNX1 (TEL-AML1) is the most common chromosomal aberration found in acute lymphoblastic leukemia in childhood. The occurrence of the ETV6-RUNX1 is associated with excellent prognosis and high sensitivity to the treatment with the enzyme L-asparaginase (ASNase). Resistance to the drug aggravates the outlook of the patient and increases the risk of treatment failure, therefore, the CLIP working group has been for a long time involved in the identification of the mechanism of action of ASNase and the origin of the resistance to it. This thesis follows previous findings of the group and is devoted to the analysis of the importance of ETV6-RUNX1 and signalization and metabolic changes accompanying shifts in the L-asparaginase resistance. In the first part of the thesis, the knockout clones with stable increased resistance to ASNase have been established thanks to the CRISPR/Cas9 system, which created frameshift in the fusion gene. The accomplishment in this regard and removal of the fusion protein was confirmed on the level of DNA, mRNA a protein expression. Nootropics and molecular mechanisms of their action

Nootropics have become the standart in the field of treating memory impairments and decline of cognition which are still more frequent problem when we consider aging of population in the western world. To better understand the capabilities and dangers of these drugs, it is necessary to widen our knowledge of the mechanism of their action. In this thesis I focus on the nootropics with the widespread use across the world (nicotine, caffeine) and on the drugs with long clinical prescription history and deeply explored effects on the cognition (methylphenidate, modafinil, piracetam, ampakines). Some categories of substances currently in the pre-clinical studies with good potential for future utilization are also briefly mentioned. The emphasis is laid mainly on summarization of the present knowledge concerning the mechanisms of action on the level of molecules, cells and the brain as a whole. The results of preclinical and clinical studies evaluating effects of these compounds on psychological values, memory and cognition are also considered here not only for CNS pathologies correction but also regarding healthy individuals.

Monitoring of raccoon dog aktivity in Opava

This work deals with invasive predator racoon dog (Nyctereutes procyonoides). It is a comprehensive work consisting of general information about its history and later spread to the whole Word, etology and biology as information collected on the basis of thousands of images and hundreds of videos from scout camera located close to natural burrow at Opava in the hunting district hunting club in Hlavnice. The emphasis here is primarily on the course of the day and night activities throughout the year, food preferences, coexistence with other kinds of beasts and ways of dealing with stressful situations. During the observation we were found that his peak of activity was among the sunrise and the second one lately after sunset. Cohabitation with badger is also possible. As for food preferences we realized that racoon dog did not prioritize any kind of food and eat everything what we gave to him.

Comparison of accounting and tax depreciation of fixed assets

Bachelor thesis is focused on accounting and tax depreciation of fixed assets, both in terms of the current czech legislation, as well as the methodology for their calculation. The first part deals with the complete theoretical knowledge necessary for the correct identification of fixed assets and the subsequent calculation of the accounting and tax depreciation. On the general practical example, which is contained in the second and third part shows the procedure for depreciation methodology.
Analysis of causes and consequences of traffic accidents in road goods conveyance

This work describes general view of a traffic accident in order to introdukce non-specific features of traffic accidents. The objective of this work is deeper understanding of this issue, of the difference between the need for improving the participants skills and the repressive measures introduced in the attempt to guarantee the observance of fundamental legal standards.
