Identification of animal hair by mass spectrometry

The goal of this bachelor work was to find out if it is possible to recognize mammal species by analyzing the proteins extracted from their hairs. This information could be of use, for example, in forensic science as evidence in cases of exotic animal species smuggling or it could be helpful for solving some specific criminal cases. Fur is a characteristic sign of mammals which is forming their body surface. It mainly consists of keratin, which is a fibrous protein. This bachelor thesis deals with proteomic analysis of animal hair using mass spectrometry working on MALDI-TOF (Matrix-Assisted Laser Desorption/Ionization - Time of Flight) principle. Analyzed animal hairs were collected from ten local domestic and wild animals, such as dog, goat, cat, fallow deer, sika deer, degu, horse, rabbit, roe deer and Belgian blue calf breed. For the analyses of samples that are rich in proteins a specific enzyme cleavage by trypsin was used. The proteins were cleaved into peptides, from which mass spectra were obtained by MALDI-TOF mass spectrometry. Then these spectra were evaluated by the Principal Component Analysis (PCA) method. Research on effectivity of outreach in physics

The thesis introduces many examples of outreach activities, primarily from the Czech Republic and other European countries. Various possibilities of measure- ments of factors of their effectivity are discussed in the thesis. The used methods were: literature review, conceptual assessment tests - pre- and post-tests (CCI, PPCI), questionnaires (IMI), data analyses of participants of activities, semi-structured interviews (for validation of concept inventories), and the work also has a glimpse of ethnography because the author has organised outreach activities for ten years. The individual chapters of the work are dedicated to: the overview of outreach activities, research methods, translation of the Calculus Concept Inventory (CCI) into Czech, the first steps for developments of the Particle Physics Concept Inven- tory (PPCI), loyalty of participants of competitions, and description of various metrics which can be used for statistical evaluation of events. 1
Analysis of sellected drugs with the use of TLC as a topic of laboratory courses

The paper aims to mediate thin film chromatography analysis by means of drugs commonly found in households (analgesic-antipyretics) to secondary school and grammar students, or to college students. For this purpose, laboratory tasks have been designed to cover all levels of the Bloom pyramid so that the educational potential is as high as possible. This is closely related to the topic of drug expiration and treatment. Pupils and students should take on both the competences of knowledge, skills, and attitudes. The analysis of application knowledge and skills of chemistry curriculum on the secondary medical schools

Key Words secondary medical school, organic compounds, work sheet, textbook, laboratory assistant, nutrition assistant, medical assistant Abstract Tuition of chemistry at medical schools is in many aspects similar to tuition at other secondary schools without focus on chemistry. Nevertheless number of lessons dedicated to chemistry rather proves that chemistry belongs to the essential subjects. The fact that this science is important for medical praxis is supported by the argument that chemistry is taught also at training colleges and universities with medical focus. The ground for the thesis is made by authors real experience gained from long-term work at Secondary Medical School and Training Medical College in Ostrava. As the main goal of the work we set to find out if the students are capable of using their knowledge, skills and habits gained by studying chemistry in the first or potentially second year of the secondary school in professional contexts, which is very often linked to nature compounds. Analysis of measurement results of concrete permeability for air and water as a basis for estimating the actual durability

Permeability surface layer of concrete is engaged in many research institutes around the world, because the surface layer is the weakest link in the entire concrete structure. This dissertation followed on the issue of global solutions and broadens the horizon of the covercrete issues. The aim of this work was to create the conversion relations between the methods for evaluating the permeability of concrete surface layer allow approximate evaluation of the surface layer of concrete depending on the current moisture. In practice mostly occur concrete, which surface mass of moisture is equal to 3 %, so this amount of moisture is selected as the reference. For comparison were used: the device TPT (Torrent Permeability Tester), device GWT (Germanns Water Permeability Tester) and methods ISAT (Initial Surface Absorption Test). A secondary aim was to create a unified methodology for measuring the device.
