Computational methods in single molecule localization microscopy Abstract Fluorescence microscopy is one of the chief tools used in biomedical research as it is a non invasive, non destructive, and highly specific imaging method. Unfortunately, an optical microscope is a diffraction limited system. Maximum achievable spatial resolution is approximately 250 nm laterally and 500 nm axially. Since most of the structures in cells researchers are interested in are smaller than that, increasing resolution is of prime importance. In recent years, several methods for imaging beyond the diffraction barrier have been developed. One of them is single molecule localization microscopy, a powerful method reported to resolve details as small as 5 nm. This approach to fluorescence microscopy is very computationally intensive. Developing methods to analyze single molecule data and to obtain super-resolution images are the topics of this thesis. In localization microscopy, a super-resolution image is reconstructed from a long sequence of conventional images of sparsely distributed single photoswitchable molecules that need to be sys- tematically localized with sub-diffraction precision. This semestral thesis describes the possibilities of source coding videos, the attributes of the videos and their recommended changes for test. Subjective methods of scores of quality and their division into metrics with references and without references are delineated. In this work, a draft of videos for test of comparison of set metrics is specified here. Further, the coding and options of attributes of videosequences for scoring of the quality of the picture is presented. The displaying system is described and quazicrowdsorcing system of collecting the datas was created.

This diploma thesis describes design and implementation of people tracking system for moving head spot light. The theoretical part describes methods of detection and tracking, practical part deals with hardware selection and software programming. DMX512 is being used to control light. Program is written in C++ and it uses OpenCV library.

This thesis deals with the development of the user interface for the application of the method Phase shifting profilometry. First deals with the theoretical approach of this method, and describes the process of image segmentation and data processing using morphological operations. In the practical part of the user interface is designed for acquiring and processing data received in Matlab.

Master´s thesis deals with methods of objects detection in the image. It contains theoretical, practical and experimental parts. Theoretical part describes image representation, the preprocessing image methods, and methods of detection and identification of objects. The practical part contains a description of the created programs and algorithms which were used in the programs. Application was created in MATLAB. The application offers intuitive graphical user interface and three different methods for the detection and identification of objects in an image. The experimental part contains a test results for an implemented program.
Test form evaluation by OCR

This thesis deals with the evaluation forms using optical character recognition. Image processing and methods used for OCR is described in the first part of thesis. In the practical part is created database of sample characters. The chosen method is based on correlation between patterns and recognized characters. The program is designed in a graphical environment MATLAB. Finally, several forms are evaluated and success rate of the proposed program is detected.
