Capillary zone electrophoresis determination of protamines
Malý, Michal ; Křížek, Tomáš (advisor) ; Kubíčková, Anna (referee)
This work deals with development and optimization of a method for separation and detection of pro- tamines using capillary zone electrophoresis. The developed method uses a fused silica capillary with inner diameter of 50 µm and effective length of 41,5 cm. Driving voltage is 30 kV. Background electrolyte is aque- ous solution of 45 mmol dm−3 phosphoric acid. Analyte is detected spectrophotometrically at wavelength of 200 nm. Sample is injected hydrodynamically. The method allows for determination of protamines in the concentration range between 11 µg ml−1 to 1000 µg ml−1 , limit of detection is 4 µg ml−1 . Viablity of the method has been verified with a real sample of NPH insulin injection. For sufficiently sensitive detection of protamines in NPH insuline it is necessary to prepare the sample in acidic environment. For the pur- poses of this work the sample was acidified by addition of background electrolyte into the sample so that the background electrolyte concentration in the resulting solution of the sample was 18 mmol dm−3 . The main advantage of the method is the rapid analysis, migration time of the analyte is about 2 min. Disadvantage of the method in comparison to alternate methods using CZE or RP-HPLC is the inability to separate individ- ual protamine peptides. This disadvantage...
Comparison of contactless conductivity detection and direct UV detection sensitivity for electrophoretic determination of N-acetylaminosaccharides
Malý, Michal ; Křížek, Tomáš (advisor) ; Kozlík, Petr (referee)
is work deals with development and optimization of a method for separation and detection of N- acetylglucosamine and its dimer and trimer using capillary zone electrophoresis with contactless conductiv- ity detection. Separation and detection of these substances is possible if sodium hydroxide is used as back- ground electrolyte. Usage of another electrolyte such as disodium phosphate leads to unacceptable baseline noise level. Optimal conditions of this method are a background electrolyte concentration of 40 mmol dm−3 and voltage of 15 kV. e method is very sensitive to background electrolyte pH, used background electrolyte also causes rapid degradation of the inner capillary wall which is why this method has proven difficult to cali- brate properly. e developed method was compared to an already existing method which uses UV detection. e existing method has lower limits of detection and quantification and is less sensitive to working condi- tions. Limits of detection and quantification of the developed method are 34 µmol dm−3 and 104 µmol dm−3 respectively. Limits of detection and quantification of the method based on UV detection are 7 µmol dm−3 and 21 µmol dm−3 respectively. Limits of quantification are that of the analyte whose detection was the least sensitive which was the dimer in both...

Economics Potential of Family Farm
Neužilová, Klára ; Malý, Michal (advisor) ; Tereza, Tereza (referee)
The aim of the diploma thesis is a study of the economic potential of a small family farm in Southern Bohemia, especially via a financial analysis, an analysis of subsidies and the SWOT method. The thesis describes the methodical basis of chosen analyses, discusses agricultural methods and current agricultural politics of the Czech Republic in the framework of the European Union. The main body of the thesis focuses on the characteristics of the studied family farm, its property and production capabilities. The financial analysis evaluates the farm based on specific indicators from year 2012 to year 2016. Furthermore an analysis of agricultural subsidies of the farm in the same time-period is carried out. Performed retrospective analyses provide an overview of the economic health of the family farm and contribute to the breakdown of the enterprise's internal environment Enterprise's internal environment is analyzed using the SWOT method, which provides four possible strategic solutions for improvement of the family farm future development. From the proposed solutions the best option is chosen based on the Multicriterial Saaty method, which is best fulfilling specified criteria. The conclusion discusses economic potential of the family farm and present and suggests possible application of the chosen strategic solution.
Modelling commodity chain of grapevine
Bílá, Iveta ; Malý, Michal (advisor) ; Tereza, Tereza (referee)
Diploma thesis deals with examining of relationships within the commodity chain of grapevine, which make a basis for prognosis creation. The first part describes methodical approaches that are needed for the creation of econometric models and also basic regularities within the commodity chain. The second part contains a summary of findings about the grapevine commodity chain. The third part introduces practical part, where forecasts based on three econometric models are created. The models are the representatives of grapevine production, wine consumption and industrial producer prices. Time series of the variables are deeply analyzed. Using the method of least squares models, estimated parameters, are founded. Afterwards they are verified on the economic, statistic and econometrics level. The verification discloses determinants of development and confirms a good prognostic characteristics thus the forecast for following three years can be created. The forecast shows slightly decreasing and finally increasing trend can be noticed, that is desired. The character of consumption model prognosis is slightly decreasing which is related to preference of domestic production and its higher price either. The prognosis of price model higlights significant fall of industrial producer prices about 24 %. The reason of that may be caused of cheaper wine imports. From domestic producers´ point of view this situation is unwelcomed.

This diploma thesis deals with the financial analysis of "Bláhův dvůr". It is a small starting young farm in the Central Bohemian Region. In the first part, I analyzed the financial analysis using financial ratios such as rate of return, liquidity, asset and debt. To determine whether a company is a profitable one, I have made the financial analysis using status indicators which are following: Altman's Z score, indices of credibility, Taffler's z-score and Quick test by Kralicek. At the end of this analysis the Cash flow is provided. The next part of my diploma thesis deals with the evaluation of the current market position of the farm using the external economic indicators such as the analysis PEST and Porter´s five forces model. After completing all the above mentioned analyses, all risks and divergences in agricultural commodities were evaluated through price trends. Using all the information, economically recoverable and revenue possibilities for future development were suggested. The main purpose of this thesis is the SWOT analysis, where I have mentioned weaknesses and threats, which will be eliminated and minimalized through business opportunities and business strengths.
