Natural products and their β-secretase inhibitory activity.
Kočová, Kateřina ; Chlebek, Jakub (advisor) ; Macáková, Kateřina (referee)
Kočová, K.: Natural compounds and their β-secretase inhibitory activity, Diploma thesis, Charles University in Prague, Faculty of Pharmacy in Hradci Králové, Department of Pharmaceutical Botany and Ecology, Hradec Králové 2015, 83 p. Data used in this Diploma Thesis have been taken from foreigner scientific literature. It provides the whole summary of natural compounds with β-secretase inhibitory activity. First chapter Alzheimer's disease briefly describes a definition, risk factors and symptomatology of the disease. Most of this chapter characterizes an etiopathology of the disease focused on amyloid theory. Finally there are also mentioned current approaches to therapy of Alzheimer's disease and those, which stay in clinical trials. Next part is dedicated to physiological functions and substrates of the β-secretase. This diploma thesis evaluates the current state of scientific research of synthetic inhibitors of β-secretase, describes the most important methods of testing inhibition and activity of β-secretase in vitro and outlines the issue of development of new inhibitors using in silico method. Main part of this work consists of the list of natural compounds with β- secretase inhibitory activity. This part is structured according to particular types of primary and secondary metabolites....

Annuity modelling in MTPL
Eštóková, Agáta ; Kočová, Karolína (advisor) ; Cipra, Tomáš (referee)
Title: Annuity modelling in MTPL Author: Bc. Agáta Eštóková Department: Department of Probability and Mathematical Statistics Supervisor: Mgr. Karolína Kočová Supervisor's e-mail address: kkocova@koop.cz Abstract: This diploma thesis focuses on the possibilities of using generational mortality tables for third party liability insurance, mainly for bodily injured. It describes the construction of generational mortality tables and the creation of RBNS claims reserves. Besides the demonstration of these models, the work analyzes the results of calculating reserves in accordance with generational mortality tables and actual mortality tables of the Czech Republic. An impor- tant element in provision calculating is the simulation of future life expectancy of the insured, i.e. random generation of life expectancy based on generational mortality data. Characteristics of the distribution of reserves are derived from simulations. Furthermore, we compare the results of stochastic and determi- nistic approach of computing the reserves. Keywords: third party liability insurance, RBNS, annuity, generational morta- lity tables.
Long-Term Insurance and Permanent Health Insurance
Kočová, Karolína ; Šváb, Jan (referee) ; Cipra, Tomáš (advisor)
This thesis deals with long-term care insurance and disability insurance. Both products are classified as products of private health insurance which is described in the first chapter. We can find characteristics of longterm care and its providers in Czech republic in the second chapter. Further, some numerical methods used in long-term care insurance are stated. The third chapter describes the British model of length of stay in institutional long-term care, the results of the study in the Great Britain are summarized in brief. The fourth chapter deals with characteristics and numerical methods of disability insurance. The fifth chapter displays the use of state model in disability insurance. The emerging costs method is illustrated on simple example. The life tables for disabled people were designed to be used in the calculation. In the conclusion we can find reasoning for the future of products mentioned above on the Czech insurance market.
Alternative methods in teaching Czech as a foreign language
Kočová, Kateřina ; Hrdlička, Milan (advisor) ; Valková, Jarmila (referee)
1 Abstract This diploma introduces three alternative methods taught in the study of foreign languages - a method of suggestopedy, Nepustil's method and the Act and Speak method. The purpose of this work is to approach the methods from a historical and methodological perspective. The main focus is aimed at inspections of alternative schools, supplied with student's questionnaires and the visual material from classes. The theoretical part discusses methods from a historical perspective, the progress and presentation of grammar and learning material. Furthermore introducing the basic concept and features of alternative methods with the posibility of applying them during the teaching of Czech language for foreigners. The practical part introduces research made up of inspections at alternative schools along with interviews with founders and successors of these methods. As analysis of the questionnaires that explore the opinions and awareness of teachers about the effectivity and usefulness of untraditional methods in teaching is also a part of it. Practical inspections and conclusions based on the results of questionnaires showed the Act and Speak method to be the most beneficial method. The practical part concludes by presenting other activities for teaching Czech for foreigners that are based on those...
Impact of pesticides on the gut microbiota of human
Kočová, Kateřina ; Rada, Vojtěch (advisor) ; Jakub, Jakub (referee)
Pesticides are substances or mixtures of substances used in agriculture against pests (animals, plants and parasitic fungi), who damage the crops, stocks of agricultural products, food and reduce livestock utility or threaten human health. The thesis consists of two parts. The first (theoretical) part describes composition and function of gut microbiota; pesticides and their classification, environmental transport and effects of these substances on human health, and glyphosate as the active substance of herbicide Roundup, its mechanism of action, environmental fate and proven effects of glyphosate on human. The aim of second (practical) part of this thesis was to test impact of pesticide Roundup on the gut microorganisms of human. The impact of pesticide was tested in in vitro pure cultures. Roundup was added in concentrations of glyphosate 17 g/L, 1,7 g/L and 0,17 g/L to the complex media and after cultivation the growth of microorganisms was evaluated. The next samples of stool were collected from human volunteers; these were cultivated similarly in the complex media with different concentrations of glyphosate. The total numbers of microorganisms, bifidobacteria, lactobacilli, enterococci and coliformn bacteria were defined after cultivation. When the pesticide was tested in pure cultures, concentration of 17 g/L glyphosate significantly inhibited total bacterial growth (P < 0.05), at lower concentrations no difference was observed. The cultivated microorganisms from samples of stool demonstrated only that bifidobacteria are sensitive to glyphosate at the highest used concentration of this pesticide compared with the control sample.
