The influence of belinostat inhibition on the activity of selected reductases from AKR and SDR superfamilies.
Slámová, Adéla ; Wsól, Vladimír (advisor) ; Macháček, Miloslav (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Bc. Adéla Slámová Supervisor: prof. Ing. Vladimír Wsól, Ph.D. Title of diploma thesis: The influence of belinostat inhibition on the activity of selected reductases from AKR and SDR superfamilies Anthracycline antibiotics (ANTs) are important antineoplastic agents. One of them, daunorubicin (DAUN), is used for the treatment of acute leukaemia and other malignancies in children and adults. Factors limiting its clinical use include mainly resistance and cardiotoxicity. Enzymes from aldo-keto reductase (AKR) and short-chain dehydrogenase/reductase (SDR) superfamilies mediated the reduction of DAUN to its C-13 alcohol metabolite daunorubicinol (DAUNOL). The metabolite is more cardiotoxic, less antineoplastic, and is causing anthracycline resistance. This diploma thesis aimed to examine the inhibitory effect of belinostat on the activity of AKR1A1, 1B1, 1B10, 1C3, and CBR1. The specific enzyme activity and inhibitory potential were estimated in vitro using recombinant enzymes, and the enzymatic production of DAUNOL was evaluated by the liquid chromatography (UHPLC) system. The inhibition was decreased in order AKR1C3  AKR1B10  AKR1A1  AKR1B1  CBR1. The most inhibited enzyme, AKR1C3, expressed 50,7%...

It deals with essential aspects not only in terms of quantity but above all the quality of the fluids we drink. Legislation regarding bottled waters and healing mineral waters is mentioned here. The following part of the work is a list of natural mineral waters and healing mineral waters that are on the market in the Czech Republic. The work continues with information on consumer behavior, including a chapter on possible health benefits for regular consumers. An important part of the work is also the evaluation of the significance of the individual minerals contained in these waters (sodium, potassium, magnesium, calcium, chlorine and fluorine) and their effects in the human body, deficiency conditions for these ions and the content of these ions in specific mineral waters. The importance of mineralization of water intake for athletes is also discussed there with more details. The practical part of the work, is in the form of a short questionnaire. The practical part tried to find out what the athletes' knowledge is about the influence of the drinking regime and the quality of...

"... The above-mentioned issues for me personally created the idea of a world in which the human race ceases to exist in the form of civilization known to us, and it is transformed back to its original collective consciousness, something that is "above" -above real physical presence, beyond basic animal processes , well above the world he created for himself, and through that killed himself. Through advanced technology and ultramodern materials, as well as minerals and vitamins, which arose from the original ancient building units, transforming the world into a new form of being. Remains of human existence in the form of countless objects and materials, which finally adapted to other forms of life (not already poisoned and weakened, but instead upgraded units) have gradually become the basic building block posthuman planetary culture. Archaeological site with an area of 510.1 million square kilometers slowly melt away in their embedded meanings and emotions seeped into polyvinyl chloride, which eventually engulfed simple cell clusters..."

OF BACHELOR THESIS Name, surname: Jana Nováková Supervisor: Ing. Adéla Slámová Consultant: Mgr. Jakub Kozel Title: Walking training within Proprioceptive neuromuscular facilitation concept in stroke patients Subtitle: Influencing of the Mid-Stance phase of stride Abstract: The topic of the bachelor's thesis is the training of walking in patients after a stroke. This acquired non-traumatic brain damage represents the second most common non-traumatic cause of death in today's modern civilization, in the Czech Republic the incidence of this disease is around 300 cases/100,000 inhabitants. The consequences of a stroke are usually numerous, and one of the resulting disorders can be hemiparetic walking with spasticity. The theoretical part includes four main chapters. The first section is devoted to the description of a commonly occurring non-traumatic brain injury, namely the description of a stroke. The second chapter describes stereotype, kinesiology and gait analysis. The third chapter includes a description of the philosophy of Proprioceptive neuromuscular facilitation (PNF), basic concepts and principles of the concept, PNF therapy of hemiparetic gait and summarizes the issue of spasticity in PNF therapy. In the last section, walking tests for patients after a stroke, their use and application in...

BACHELOR THESIS ABSTRACT Name, surname: Václav Matys Supervisor: Ing. Adéla Slámová Consultant: Ing. Mgr. Jana Horynová Title: Stimulation of swallowing within PNF concept in pacient with acquired brain injury A swallowing disorder often occurs as a complication in acquired brain injury diagnosis. In more serious cases it can constitute even life-threatening condition, primarily due to the risk of aspiration and possilbe subsequent development of an inflammatory lung disease. This thesis deals with a possibility of therapeutic influencing of neurogenic dysphagia with utilization of elements of the concept of proprioceptive neuromuscular facilitation - PNF. Contrary to the fact that exercise on the PNF basis to influence dysphagia isn't used frequently in clinical practice, it represents highly effective and for the therapist easily managable method of therapy. The practical part contains a case history of patients with dysphagia, to whom the therapy on the PNF basis was applied. The goal of the bachelor thesis is to create an educative material for a community of experts, particularly physiotherapist as well as speech pathologists or occupational therapists, and thus support interdisciplinary cooperation in the care of dysphagia patients. The result of the thesis is a formation of the therapeutic procedure...
