Vyhledávání jaderného transportinu faktoru ISW1 remodelujícího chromatin u Saccharomyces cerevisiae

Several intragenously-tagged versions of the ISW1 gene were constructed to follow Isw1p localization either in living (GFP-fusions) or in fixed cells (c-Myc-fusions). The whole set of available importin mutants was transformed with vectors carrying an intragenously-tagged ISW1 gene. In the strains where the importins Kap120p, Nmd5p, Sxm1p, Kap114p, Los1p, Msn5p, Kap123p and Pdr6 were deleted, the Isw1p-GFP fusion regulated by the ISW1 promoter was localized always in the nucleus. This suggests that none from these importins is required for nuclear localization of Isw1p. The localization of the Isw1p-GFP in the mtr10-1 strain differed from that observed in all the other analyzed importin mutants. Our results indicate that Mtr10p could be a good candidate for Isw1p importin.
Analysis of the catechol degradation operon from Rhodococcus erythropolis

Three genes of phenol-degrader Rhodococcus erythropolis involved in degradation of catechol and one gene coding for their activator were isolated and characterized.The obtained knowledge will contribute to construction of strains with more efficient phenol degradation.
Bioavailability of PAH and its effects on biodegradation during composting

The publication deals with application of composting process on remediation of soils contaminated with PAHs. Bioavailability of PAHs was tested with sequential SFE and the data were compared with the results of remediation. SFE is suitable to predict results of bioremediation.

Altered tight junctions (TJs) structure was detect histochemically in colitic mice using antibodies against ZO-1 proteins. Structural changes of TJs was important feature of intestinal inflammation

Metabolic fluxes in pathways of biosynthesis of branched-chain amino acids can be modulated by different changes in expression of genes of the desired pathway as well as those of competing pathways. Site-directed mutagenesis of promoters of the respective genes is a suitable tool to obtain desired changes in gene expression.

We compared influence of hydrolytically and enzymatically cleavable conjugates based on N-(2-hydroxypropyl)methacrylamide on proliferatin activity and expression of mRNA of genes involved in apoptotic signalling pathway. We found elevated expression of Fas and bax mRNA in cells incubated with hydrolytically cleavable conjugates.
