Abstract: This study explores potential inter-subunit interactions of the hexameric Orai1 channel. Our investigation include the loss of function (LoF) mutants Orai1 K85E and E149K residues. We demonstrated that the Orai1 K85E mutation reduces homomerization via fluorescence resonance energy transfer (FRET) measurements, while the homomerization of E149k remains comparable to the Orai1 wild type In addition, we identified spatial proximity of Orai1 V181 and C143 by implementing the proximity-dependent chemical crosslinking unnatural amino acid (UAA) O-(2-bromoethyl)-tyrosine (BrEtY). Overall, we provide information about the structural assembly of the Orai1 channel.
