Guanine quadruplexes are non-canonical secondary structures of nucleic acids that can form under suitable conditions in guanine-rich sections of DNA and RNA. This ba- chelor's thesis deals with similar structures, the mononucleotide guanine quadruplexes (mG4), which form at a sufficient concentration by self-assembly of free guanine mono- nucleotides. The subject of investigation is the effect of proton-deuterium exchange on the structure and stability of mG4 depending on the type of charge compensating central cation or the presence of ribose vs. 2'-deoxyribose. Using Raman spectroscopy, samples are studied in liquid phase, in the form of an amorphous lyophilisate and using the me- thod of drop-applied coatings. Using simple association models, the melting temperature of mG4 aggregates in a heavy water environment is determined. 1 Guanínové kvadruplexy sú nekanonické sekundárne štruktúry nukleových kyselín, ktoré pri vhodných podmienkach vznikajú v častiach DNA a RNA bohatých na guanín. Táto bakalárska práca sa venuje obdobným štruktúram, tzv. mononukleotidovým guaní- novým kvadruplexom (mG4), ktoré pri dostatočnej koncentrácii vznikajú samoasociáciou voľných guanínových mononukleotidov. Predmetom skúmania je vplyv protón-deutériovej výmeny na štruktúru a stabilitu mG4 v závislosti na druhu náboj kompenzujúceho cen- trálneho katiónu alebo prítomnosti ribózy či 2'-deoxyribózy. Pomocou Ramanovej spek- troskopie sú študované vzorky v kvapalnej fáze, vo forme amorfného lyofilizátu a využitím metódy kvapkovo nanášaných povlakov. Prostredníctvom jednoduchých asociačných mo- delov je stanovená teplota topenia agregátov mG4 v prostredí ťažkej vody. 1
