Lidský selenoprotein H (SelH) je důležitý pro vývoj, reakci na oxidační stres a mitochondriální biogenezi. Jako protein obsahující selenocystein (Sec, U) má SelH vysoce konzervovaný motiv CXXU, který je konzistentní s jeho rolí v redoxní regulaci. Ačkoli se proteiny obsahující Sec nevyskytují u suchozemských rostlin, dva blízce příbuzné homology SelH byly dříve identifikovány u Arabidopsis thaliana (jmenovitě AtSelH1 a AtSelH2). Oba jsou jaderné proteiny s neznámými funkcemi, které obsahují konzervovaný motiv CXXC, ve kterém je Sec nahrazen cysteinem. Abychom porozuměli jejich molekulární funkci, použili jsme transgenní rostliny Arabidopsis nadměrně exprimující AtSelH1 a AtSelH2 a pozorovali jsme silný účinek jejich nadměrné exprese na růst a vývoj, který vedl k poškození listů. Dále jsme sledovali subcelulární lokalizaci AtSelH1 a AtSelH2 v centromerických oblastech a jadérku. Při hledání funkcí domnělých proteinových interaktorů AtSelH1 a AtSelH2 jsme pozorovali četné zastoupení proteinů zapojených do biogeneze ribozomů, což naznačuje funkční roli AtSelH1 a AtSelH2 v tomto procesu. Toto jsou první vodítka k pochopení funkce AtSelH1 a AtSelH2. Human selenoprotein H (SelH) is significant for development, response to oxidative stress, and mitochondrial biogenesis. As a selenocysteine (Sec, U)-containing protein SelH has a highly conserved CXXU motif consistent with its role in redox regulation. Although Sec-containing proteins do not occur in terrestrial plants, two closely related SelH homologs have been previously identified in Arabidopsis thaliana (namely AtSe-lH1 and AtSelH2). Both are nuclear proteins with unknown functions that contain a conserved CXXC motif in which Sec is substituted by cysteine. To understand their molecular function, we used transgenic Arabidopsis plants overexpressing AtSelH1 and AtSelH2, and observed strong effect of their overexpression on growth and develop-ment, that ultimatley resulted in leaf lesions. We further monitored the subcellular localization of AtSelH1 and AtSelH2 in centromeric regions and nucleolus. By assessing the roles of their putative protein interactors, we observed functional enrichment of proteins implicated in ribosome biogenesis suggesting a functional role of AtSelH1 and AtSelH2 in this process. These are the first clues towards understanding of the function roles of AtSelH1 and AtSelH2.
