[cze][eng] Polynenasycené mastné kyseliny (PUFA) n-3 mají pozitivní vliv na hojení ran, ale studie s nimi produkují nekonzistentní výsledky. V této diplomové práci byla pilotně využita hmotnostní zobrazovací spektrometrie s laserovou desorpcí/ionizací za účasti matrice s průletovým analyzátorem (MALDI-TOF MSI), a byla optimalizována pro hodnocení dietárního vlivu olejů na hojení kožních ran u potkanů. Pro studium byly využity: rybí olej; palmový olej; olej extrahovaný z mikrořasy Schizochytrium sp. a světlice barvířské. Tyto jednotlivé oleje se liší obsahem PUFA n-3 a n-6. Dále byl studován dietární vliv těchto olejů pomocí imunohistochemických analýz, a chromatografie pro určení obsahu hydroxyprolinu. Díky úspěšné aplikaci všech výše zmíněných metod byl komplexně prokázán rozdílný účinek studovaných olejů na hojení ran u modelového zvířete potkana.Polyunsaturated fatty acids (PUFA) n-3 have positive effect on a wound healing but studies with them produce inconsistent results. In this thesis, the matrix-assisted laser desorption/ionization time-of-flight mass spectrometry imaging (MALDI-TOF MSI) was utilized and optimized for evaluation of effect of dietary oils on cutaneous wound healing in the rat model. For studies were used: fish oil; palm oil; Schizochytrium microalga oil and safflower oil. This individual oils differentiates in PUFA n-3 and n-6 content. Further, effects of these dietary oils were studied using immunohistochemical analyses, and chromatography for hydroxyproline determination. Thanks to the successful application of all of the above mentioned methods, a different effect of the studied oils on wound healing in the rat model was comprehensively demonstrated.
