Abstract: The Master Thesis on the topic “Detection and identification of possible mycoviruses within Diaporthe sp., a major pathogen of Czech grapevines” was compiled in 2017 – 2018 at laboratory of Department of Forest Protection and Wildlife Management, Faculty of Forestry and Wood Technology and Department of Genetics, Faculty of Horticulture, Mendel University in Brno. The thesis was focused on detection the presence of dsRNA in isolates of Diaporthe sp., a pathogen of grapevines. An Output of thesis was confirmation of presence dsRNA in length of 2 – 2.5 kb, which could correspond to mycoviral family Narnaviridae, respectively to the genus Mitovirus. Used methods are based on isolation and preparation of fungal cultures on medium, followed by extraction of dsRNA according to protocol modified by Morrison and Dodds (1979). The procedure described in this protocol is based on binding the nucleic acids to cellulose powder in 15% ethanol buffer in columns. For the identification of found dsRNA particles is necessary to continue the investigation by sequencing.
