[cze][eng] Tato bakalářská práce se zabývá problematikou identifikace genu VRN-H1 a VRN-H2 a jejich vlivu na růstový typ ječmene. Součástí práce byla sumarizace možností detekce různých alel těchto genů. V rámci experimentální části byly testovány výskyty alel genu VRN-H1 a VRN-H2 u vybraných odrůd ječmene, které se lišily požadavky na vernalizaci. U ozimých odrůd ječmene byl potvrzen výskyt ozimých alel v obou systémech genů VRN-H1 a VRN-H2, zatímco u jarních odrůd byl stanoven výskyt jarních alel. Odrůda Tadmor nemá jednoznačně určený vegetační typ, ale byla u ní zjištěna jarní alela v genu VRN-H1 a ozimé alely v genu VRN-H2. Následně byla ověřena závislost regulace exprese genu VRN-H1 u ozimých odrůd ječmene na vývojové fázi dané stádiem vývoje růstového vrcholu dle Waddingtonovy stupnice. Bylo prokázáno výrazné zvýšení (50 x) úrovně exprese tohoto genu u rostlin, které se se nacházely na počátku generativní fáze (W4,5–5,0) oproti rostlinám, u kterých ještě nebyla ukončena vernalizace a nacházely se ve vegetativní fázi (W1,5–1,9).This bachelor work deals with the identification of VRN-H1 and VRN-H2 genes and their influence on barley's growth habit. Part of the work is devoted to summarization of possibilities for detection different alleles of these genes. In experimental part in selected barley varieties, which differed by their growth habit, alleles at the VRN-H1 and VRN-H2 genes were tested. In winter barley varieties, the occurrence of winter alleles in both VRN-H1 and VRN-H2 gene systems was confirmed. While the occurrence of spring alleles was determined in spring varieties. The growth habit of the Tadmor variety was not unambiguous, but according to genotype the spring allele was determined at the VRN-H1 gene and the winter allele was determined at the VRN-H2 gene. Subsequently, the regulation of the VRN-H1 gene expression in winter barley varieties depending on the developmental phase was verified according to the Waddington scale. The developmental phase was assessed by the stage of root apex development. There was a significant increase (50x) in VRN-H1 gene expression in plants which were at the beginning of the generative phase (W4.5-5.0) compared to plants which had not yet completed vernalization and were in the vegetative phase (W1, 5–1.9).
