RNDr. Lenka Skálová, Ph.D. Název diplomové práce: Testování účinnosti a toxicity nových potenciálních anthelmintik OMK3 a OMK4 Haemonchus contortus (vlasovka slezová) je parazitem menších přežvýkavců, hlavně ovcí a koz. Způsobuje onemocnění zvané haemonchóza, jehož hlavním příznakem je anémie, která může skončit až smrtí zvířete. Pro léčbu tohoto onemocnění se používají anthelmintika. Tato léčiva ale přestávají být účinná, protože na ně vzniká rezistence. Haemonchóza se proto stává velkým celosvětovým problémem. Z toho důvodu je nutné hledat nová anthelmintika s novým mechanismem účinku. V této práci byly zkoumány potenciální anthelmintiké účinky nových derivátů kyseliny benzhydroxamové, OMK3 a OMK4. Cílem práce bylo zjistit, jestli tyto látky mají účinek na vajíčka, larvy a dospělce H. contortus, porovnat jejich účinek na citlivém a rezistentním kmeni H. contortus a testovat jejich potenciální toxicitu na játrech ovcí. K těmto účelům byla izolována vajíčka, larvy a dospělci H. contortus, připraveny precizní tkáňové řezy z ovčích jater a použity testy: líhnutí vajíček, sledování motility a stanovení koncentrace ATP bioluminiscenční metodou. It causes a disease called haemonchosis, the main symptom of which is anemia, whitch can lead to the death of the animal. Anthelmintics are used to treat this disease. However, these drugs have become ineffective due to resistance developing against them. Haemonchosis is thus becoming a significant global problem. Therefore, it is necessary to search for new anthelmintics with a new mechanism of action. In this study, the potential anthelmintic effects of new derivates of benzhydroxamic acide, OMK3 and OMK4, were investigated. The aim of this study was to determine whether these substances have an effect on eggs, larvae and adults of H. contortus, and to compare their effects on sensitive and resistant strains of H. contortus and to test their potential toxicity on sheep livers.
