Terpenoidy jsou nejbohatší skupinou přírodních látek, které se vyskytují v podobě desítek tisíc rozmanitých molekul napříč různými formami života, od mikroorganismů přes rostliny až po živočichy. Terpenoidy nalézáme také u hmyzu, druhově nejpočetnější živočišné skupiny, kde zastávají úlohu zejména v komunikaci a chemické obraně. Ve srovnání s bohatými znalostmi o biosyntéze terpenoidůu rostlin jsou naše vědomosti o biosyntetickém původuterpenoidů u hmyzu velmi kusé a pocházejí vesměs z výzkumu posledních několika let. Tyto nové poznatky ukazují, že schopnost biosyntézy terpenoidů se vyvinula opakovaně a nezávisle u jednotlivých vývojových linií hmyzu a že hmyzí terpensynthasy vznikají prostřednictvím neofunkcionalizace genů, které byly původně zodpovědné za produkci isoprenylpyrofosfátů, a jejich následnou funkční diverzifikací. Mezi ostatními skupinami hmyzu vyčnívají termiti, kteří využívají širokou škálu isoprenoidů pro komunikaci a unikátní chemickou obranu. Nicméně vznik terpenoidních látek u termitů je stále neznámý. Tato práce se zabývá funkční charakterizaci tří domnělých terpensynthas termita druhu Nasutitermes takasagoensis. Exprese vybraných proteinů probíhala ve dvou bakteriálních systémech a pro jejich purifikaci byla použita metoda afinitní chromatografie na vázaných kovových iontech.... Terpenoids are natural compounds, which can be found in the form of tens of thousands diverse molecules in all forms of life. Terpenoids are mostly represented in plants, microorganisms,and animals. It should not be a surprise that terpenoids are often found in insects, the most species-rich class of Metazoa. In insects terpenoids are mostly used for communication as pheromones and for chemical defence. In comparison to widespread knowledge of terpene (and terpenoid) biosynthesis in plants we lack thorough knowledge in insects. Studies conducted in past few years showedthat the ability to synthesize terpenoids has occurred independently and repeatedly in various lineages of insects. It was also found that terpene synthase genes evolved via duplication and neofunctionalization of specific genes, which were responsible for isoprenyldiphospate synthesis. Despite these discoveries the terpenoid synthesis in insects is mostly unknown and remains to be elucidated. This thesis aims to functionally characterize three candidate terpene synthases from the termite Nasutitermes takasagoensis. Expression of selected proteins was done in two bacterial systems.
