Tématem bakalářské práce je příprava neurosteroidních proléčiv za účelem optimalizace jejich fyzikálně-chemických a metabolických vlastností s cílem usnadnit jejich transport membránami, zejména průchod přes hematoencefalickou bariéru. Neurosteroidy jsou endogenní látky, které vznikají v mozku z cholesterolu. Jejich charakteristickou funkční skupinou je hydroxylový substituent v poloze C-3. Tento substituent je významným strukturním rysem podmiňujícím biologickou aktivitu (např. alosterickou modulaci receptorů kyseliny - aminomáselné). Je však zároveň velmi snadno metabolizován a volná hydroxylová skupina v molekule neurosteroidu zvyšuje její hydrofilicitu a prostup membránou omezuje. V průběhu práce budou připraveny: 1) neurosteroidy s různými skelety jako výchozí látky a 2) analogy neurosteroidů s esterovou skupinou v poloze 3 jako proléčiva. Substituenty byly vybrány na základě literární rešerše esterových proléčiv ovlivňujících průnik přes hematoencefalickou bariéru. Pro nově připravené látky budou stanoveny jejich metabolické a fyzikálně-chemické vlastnosti, jako například rozpustnost, permeabilita a metabolická stabilita v rámci spolupráce na Ústavu organické chemie a biochemie, Akademie věd České republiky. Klíčová slova: neurosteroidy, proléčiva, permeabilita, stabilita This bachelor thesis explores various methods for neurosteroid prodrugs synthesis. The thesis aims to optimize the physicochemical and metabolic properties of neurosteroids to facilitate their transport across membranes, in particular the permeability of the blood-brain barrier. Neurosteroids are endogenous compounds that are synthesized in the brain from cholesterol. Their characteristic functional group is the hydroxyl substituent at the C-3 position. This substituent is a significant structural feature for biological activity (e.g., allosteric modulation of γ-aminobutyric acid receptors). However, the C-3 hydroxyl group can be easily metabolized. Moreover, the free hydroxyl group in the neurosteroid molecule increases its hydrophilicity which can limit the transport across the membrane. Therefore, within this thesis: 1) neurosteroids with different skeletons and 2) analogs of neurosteroids with ester group in C-3 position, will be prepared. The C-3 substituents have been selected based on literature research identifying ester prodrugs targeting improved blood-brain barrier permeability.
