6 ABSTRAKT Univerzita Karlova, Farmaceutická fakulta v Hradci Králové Katedra biologických a lékařských věd Název diplomové práce: Vliv glipizidu na expresi a funkci endoglinu a sním souvisejících biomarkerů endotelové dysfunkce u diabetu mellitu II.typu Autor: Iva Vochozková Vedoucí diplomové práce: PharmDr. Katarína Tripská, Ph.D. Cíl práce: Cílem této diplomové práce bylo zjistit, jak glipizid ovlivňuje expresi a funkci endoglinu, jeho transkripčních faktorů a souvisejících biomarkerů endotelové dysfunkce, hladinu solubilního endoglinu a adhezi monocytů na lidské diabetické endotelové buňky koronárních artérií. Metody: V této diplomové práci jsme pracovali s lidskými diabetickými endotelovými buňkami koronárních artérií. Ty jsme v různých časech a koncentracích premedikovali glipizidem. Pomocí průtokové cytometrie jsme měřili proteinovou expresi endoglinu a biomarkerů endotelové dysfunkce. Pomocí průtokové cytometrie jsme měřili také adhezi monocytů k endotelovým buňkám. ELISA metodou jsme detekovali hladiny solubilního endoglinu a pomocí PCR jsme měřili mRNA expresi endoglinu, jeho transkripčních faktorů a markerů zánětu. Výsledky: Prokázali jsme, že premedikace 200µM koncentrací glipizidu vedla ke snížení proteinové exprese endoglinu po 24 i 48 hodinách. Po 24 hodinách došlo taktéž ke zvýšení... 8 ABSTRACT Charles University, Faculty of Pharmacy in Hradec Kralové Department of Biological and Medical Sciences Title of Diploma Thesis: Effect of glipizide on the expression and function of endoglin and related biomarkers of endothelial dysfunction in type II diabetes mellitus Author: Iva Vochozková Supervisor of Diploma Thesis: PharmDr. Katarína Tripská, Ph.D. Aim: The aim of this thesis was to determine how glipizide affects the expression and function of endoglin, its transcription factors and related biomarkers of endothelial dysfunction, soluble endoglin levels and monocyte adhesion to human diabetic coronary artery endothelial cells. Methods: In this thesis we worked with human diabetic coronary artery endothelial cells. These were premedicated with glipizide at various times and concentrations. We measured protein expression of endoglin and biomarkers of endothelial dysfunction using flow cytometry. We also measured monocyte adhesion to endothelial cells using flow cytometry. We detected soluble endoglin levels by ELISA and measured mRNA expression of endoglin, its transcription factors and markers of inflammation by PCR.
