[cze][eng] Progrese buněčného cyklu je složitě regulována mnoha proteiny. Mezi těmito proteiny hrají klíčovou roli inhibitory cyklin-dependentních kináz (CKI), které jsou tradičně spojované s inhibicí cyklin-dependentních kináz (CDK) a zastavením buněčného cyklu. Nicméně nedávný výzkum odhaluje, že jejich funkce je v kontextu rakoviny mnohem složitější. CKI inhibitory nejsou v mnoha typech nádorů vůbec mutovány a jejich exprese je zachována, nebo dokonce zvýšena, což naznačuje další mechanismy, kterými ovlivňují patogenezi nádorů. Zásadní vliv na jejich funkci mají posttranslační modifikace, zejména fosforylace, které ovlivňují jejich lokalizaci a stabilitu. Tyto modifikace pak mohou mít například vliv na buněčnou motilitu, apoptózu nebo opravu DNA. To může CKI přeměňovat z tumor supresorů na tumor promotory, nebo naopak posilovat jejich tumor supresorní vlastnosti. Among these proteins, cyclin- dependent kinase inhibitors (CKIs) play a key role, traditionally associated with cyclin- dependent kinase (CDK) inhibition and cell cycle arrest. However, recent research reveals that their function is much more complex in the context of cancer. CKI inhibitors are not mutated at all in many tumor types, and their expression is maintained or even increased, suggesting additional mechanisms by which they influence tumor pathogenesis. Post-translational modifications, especially phosphorylation, which affect their localization and stability, have a fundamental influence on their function. These modifications can then, for example, affect cell motility, apoptosis, or DNA repair. This can convert CKIs from tumor suppressors to tumor promoters or vice versa, strengthening their tumor suppressor properties.
