Diplomová práce se zabývá vyhodnocením směrového uspořádání kolagenních vláken ve společné krční tepně a jejich vlivem na její napjatost a deformaci. První, rešeršní, část práce obsahuje popis tepenné stěny, přehled některých hyperelastických konstitutivních modelů využívaných pro její modelaci a shrnutí současného stavu poznání z oblasti metod používaných pro vyšetřování organizace kolagenu v krčních tepnách. Druhá, experimentální, část práce prezentuje výsledky získané z měření orientace kolagenu v lidských společných karotidách, které bylo autorkou provedenou za pomocí světelné polarizační mikroskopie a automatického algoritmu. Z naměřených dat je zřejmé, že ve stěně tepny existuje jedna rodina obvodově orientovaných vláken, což vyvrací zavedenou hypotézu o dvou symetrických rodinách, které je uváděno ve světové literatuře. Předmětem třetí části práce byla aproximace křivek naměřených při ekvibiaxiální tahové zkoušce krkavice pomocí modelu Yeoh a modifikovaných modelů HGO a GOH, u kterých byla izotropní část tvořená modelem Neo-Hooke nahrazená tříparametrickým modelem Yeoh, který je schopen popsat zpevnění tkáně. Poslední, výpočtová, část práce využívá konstanty popisující použité konstitutivní modely a strukturní parametry získané z mikroskopického měření pro tvorbu modelu materiálu, který byl následně využit pro simulaci tepny zatížené středním arteriálním tlakem a zvýšeným systolickým tlakem při dvou velikostech axiálního předpětí typických pro jinou věkovou skupinu. Implementací obvodově orientovaných vláken do výpočtového modelu dochází ke snížení celkové deformace tepny, zvýšení obvodového a snížení axiálního napětí. The diploma thesis deals with the evaluation of the directional arrangement of collagen fibres in the common carotid artery and their influence on its stress and deformation. The first part of the thesis, literature search, contains a description of the arterial wall, a review of some hyperelastic constitutive models used for its modelling and a summary of the current state of knowledge in the field of methods used for the investigation of collagen organisation in carotid arteries. The second, experimental, part of the paper presents results obtained from measurements of collagen orientation in human common carotid arteries, which were obtained by the author using polarized light microscopy and an automated algorithm. From the measured data, it is clear that there is one family of circumferentially oriented fibres in the artery wall, which disproves the acknowledged hypothesis of two symmetric families reported in the world literature. The subject of the third part of the thesis was the approximation of the curves during the equibiaxial tensile testing of the carotid artery using the Yeoh model and modified HGO and GOH models, in which the isotropic part formed by the Neo-Hooke model was replaced by the three-parameter Yeoh model, which is able to describe the tissue stiffening. The last, computational, part of the work uses the constants describing the used constitutive models and the structural parameters obtained from microscopic measurements to create a model of material, which was then used to simulate an artery loaded with mean arterial pressure and elevated systolic pressure at two magnitudes of axial prestrain typical of different age groups. The implementation of circumferentially oriented fibres in the computational model reduces the overall deformation of the artery, increases the circumferential and reduces the axial stress.
