Cílem této práce byly syntézy ve vodě rozpustných kvantových teček (QDs) pomocí různých prekurzorů a stabilizátorů a zjištění toxicity syntetizovaných QDs nutných pro in-vivo zobrazování. Tato práce je zaměřena na syntézy ve vodě rozpustných QDs (MPA-CdTe, MPA-CdTe/ZnS, MSA-CdTe, MSA-CdTe/ZnS, GSH-CdTe, GSH-CdTe/ZnS, TGA-CdTe, TGA-CdTe/ZnS, GSH-ZnSe a GSH-ZnSe/ZnS) a zejména na jejich toxicitu. Syntetizované QDS vyznačující se vysokou intenzitou fluorescence a záporným zeta potenciálem byly vybrány vzhledem k jejich vhodnosti pro tuto práci. Výsledné hodnoty QDs byly vyhodnoceny a u QDs s optimálními vlastnostmi (maximální intenzita, FWHM, zeta potenciál) byla dále studovaná toxicita. Toxicita QDs byla určena pomocí MTT testu na buněčné linii HEK 293, při kterém bylo podle rešerše potvrzeno, že QDs se strukturou jádro/obal jsou méně toxické než struktura jádro. Z výsledků vyplývá, že toxicita námi syntetizovaných QDs je nejnižší u MPA-CdTe (struktura jádro) a MPA-CdTe/ZnS (struktura jádro/obal). The aim of this work was to synthesise water-soluble QDs using different precursors and stabilizers and to determine the toxicity of the synthesized QDs by in vivo imaging. Experiments were performed on water-soluble QDs (MPA-CdTe, MPA-CdTe/ZnS, MSA-CdTe, MSA-CdTe/ZnS, GSH-CdTe, GSH-CdTe/ZnS, TGA-CdTe, TGA-CdTe/ZnS, GSH-ZnSe and GSH-ZnSe/ZnS ) and toxicity was measured. Synthesized QDs were characterized by high intensity (fluorescence spectroscopy), FWHM and zeta potential (ZS Zetasizer) were selected due to their suitability for this task. The toxicity of QDs was determined by the MTT assay on the cell line HEK 293. The experiments show that a core/shell structure is less toxic than a core structure. The results indicate that the toxicity of our synthesized QDs is the lowest for MPA-CdTe (core structure) and MPA-CdTe/ZnS (core/shell structure).
