Extracelulární vesikuly jsou poměrně nově objeveným způsobem mezibuněčné komunikace. Problematika těchto membránou ohraničených váčků je stále značně neprobádaná, a to především na poli jejich role při nádorových onemocněních. Cílem této pilotní studie bylo pokusit se detekovat platinu ve vesikulech pomocí ICP-MS. Pro tento účel byly použity vzorky z buněčných linií nádorů ovarií, které byly vystaveny platinovým derivátům (typ chemoterapeutika). Tato metoda se ukázala jako schopná nejen platinu detekovat, ale i spolehlivě stanovit její koncentraci. Množství platiny ve vesikulech se pohybovalo kolem 1–2 % z celkově dodanému množství platiny do systému. Výsledky se lišily dle použitého platinového derivátu, buněčné linie a počtu buněk, kterými byly vesikuly uvolňovány. Nejvíce platiny bylo stanoveno ve vesikulech linie SK-OV-3, která je na tuto léčbu přirozeně rezistentní, a to u všech platinových derivátů. Využití ICP-MS je tedy vhodné i pro další výzkum této problematiky. Extracellular vesicles are a newly discovered way of cell-to-cell communication. The issue is still significantly unexplored, especially in the case of cancer. The goal of this pilot study was to attempt to detect platinum contained in extracellular vesicles by ICP-MS. In this study, samples of ovarian cancer cell lines treated with platinum derivatives, a type of chemotherapeutic drug, were used. This method was proved as able to detect platinum, even as able to determine its concentration reliably. The quantity of platinum in vesicles was around 1–2 % of the total platinum added to the system. The results varied according to the used platinum derivative, the cell line and the number of cells releasing the vesicles. Most platinum was determined in vesicles of the SK-OV-3 line, which is naturally resistant to this treatment, for all platinum derivatives. From these results it can be concluded that the use of ICP-MS is also advisable for additional research on this issue.
