Bakalářská práce se věnuje vlastnostem lidských tkání a bakterií z mikroskopického hlediska. Dále popisuje fyzikální principy mikroskopovacích technik SNOM a elektronové mikroskopie (TEM, SEM, ESEM) a jejich vhodnost pro pozorování biologických preparátů. Následující část se zabývá transformací dat z elektronového mikroskopu z časové do obrazové roviny a práci se vzniklými obrazy. Další kapitola je věnována experimentální přípravě preparátů pro SEM/ESEM a SNOM. Protože jsou zde v současné době tendence využít mikroskopovací techniku SNOM v biologické výzkumu je cílem práce srovnání možností zmíněných mikroskopovacích technik pro pozorování biologických vzorků a zhodnocení, zda je možné pro tyto účely nahradit obecně drahý EM levnějším SNOM. EM i SNOM totiž umožňují získat rozdílnou nicméně srovnatelnou a komplementární informaci o zkoumaném vzorku.
Bachelor thesis deals with characterization of human tissues and bacteria from the microscopic point of view. Physical principles of SNOM and electron microscopy (TEM, SEM, ESEM), together with their suitability for observing of biological specimens, were presented. In the next section, there is focus on data transformation from electron microscopy from time plane to image plane and their following processing. Next section is about experimental preparation of specimens for observing in SEM, ESEM and SNOM. Currently there is an idea to use scanning near-field optical microscope for biological research. That is why the goal of this thesis is compare SNOM and EM techniques, mentioned above. In this thesis we try to use SNOM as a cheaper option than EM in the observation of biological specimens. EM and SNOM allow obtaining different, however comparable and complementary information of the specimen.
