[cze][eng] Muskarinové receptory patří do skupiny receptorů spřažených s G-proteiny. G-proteiny jsou heterotrimerické proteiny vázající GTP předávající signál od receptorů k efektorům, ty pak mění hladinu druhých poslů v buňce. Jednotlivé podtypy muskarinových receptorů se spřahují s různými typy G-proteinů a dle toho vyvolávají různé změny v hladinách druhých poslů. Jednotlivé podtypy muskarinových receptorů se dále liší lokalizací a funkcí, kterou zastávají. Podtyp muskarinového receptoru M3 se nachází převážně na periferii, kde zprostředkovává kontrakci hladké svaloviny a sekreci žláz s vnitřní a vnější sekrecí. Cílem této bakalářské práce bylo popsat tkáňově specifické signální dráhy M3 receptoru a jeho fyziologické role jak na periferii, tak i v centrálním nervovém systému. Dále je popsána úloha M3 receptorů v řadě patologií a uvedeno možné uplatnění M3 receptorů jako farmakologického cíle. G-proteins are heterotrimeric GTP-binding proteins that transfer signals from receptors to effectors. Effectors change levels of second messengers in the cell. Individual subtypes of muscarinic receptors bind to various classes of G-proteins. According to the G-protein coupling, muscarinic receptors change levels of various second messengers. Individual subtypes of the muscarinic receptor also differ in location and function. The M3 muscarinic receptor subtype is located primarily on the periphery where it mediates smooth muscle contraction and endocrine and exocrine secretion. The goal of this bachelor thesis was to describe the tissue-specific signalling pathways of the M3 receptor and their physiological roles in the periphery as well as in the central nervous system. Further, the role of M3 receptors in several pathologies is described. Finally, the M3 receptors as a possible pharmacological target will be discussed.
