[cze][eng] Rod Wolffia jsou vodní rostliny s extrémně redukovanou morfologií a rychlým reprodukčním cyklem s nejkratší zaznamenanou dobou zdvojení méně než 30 hodin. Tyto nejmenší známé kvetoucí rostliny jsou vynikajícím modelem pro biotechnologie i pro studium molekulárních mechanismů růstu a vývoje. Teoretická část této práce představuje rod Wolffia a diskutuje očekávané budoucí aplikace. Experimentální část je věnována charakterizaci proteomu W. australiana s využitím dvou komplementárních strategií pro frakcionaci proteomu pomocí C18 off-line frakcionace peptidů a online frakcionační techniky založené na iontové mobilitě. Následující část popisuje analýzu proteomu W. australiana pěstované v bohatém živném médiu. Nakonec jsou představeny výsledky profilování proteomu a metabolomu W. australiana v reakci na ionty kadmia.Wolffia spp. are aquatic plants with extremely reduced morphology and rapid reproduction cycle with the shortest recorded doubling time of under 30 hours. These smallest known flowering plants are thus excellent for biotechnology and studying molecular mechanisms underlying growth and development. The theoretical part of this thesis reviews Wolffia spp. and discusses prospective future applications. The experimental part is dedicated to the characterization of W. australiana proteome, employing two complementary strategies for proteome fractionation using an off-line C18 peptides fractionation and an online ion mobility-based fractionation technique. That part is followed by a proteome analysis of W. australiana grown in nutrient-rich media. Finally, results of proteome and metabolome profiling of W. australiana in response to cadmium ions are presented.
